Objective:To investigate potential effects of some natural drugs on pharmacokinetics of anti-AIDS chemical drugs.Methods:in the present study,the chemical drugs included Didanosine(ddI),Stavudine (d4T),Nevirapine(NVP),Atazanzvir(ATV)and Indinavir(IDV),while the natural drugs included Tangcao tablet(TC)and Wuzhi capsule(WZ).Self-control method and parallel-control method were applied.①Effects of TC on pharmacokinetics of ddI+d4T+NVP studyTen rats were divided into two groups,respectively:control group,p,o.administrated ddI+d4T+NVP,TC group,p.o.administrated ddI+d4T+NVP and TC.②Effects of WZ on pharmacokinetics of ATV studyFive rats and four beagles were used.ATV was single p.o.administrated before and after the 7-day WZ successive p.o.administration,respectively.③Effects of WZ on pharmacokinetics of IDV studySix beagles were used.IDV was single p.o.administrated before and after the 7-day WZ successive p.o.administration.The concentrations of ddI,d4T and NVP in plasma were measured by HPLC,and the concentrations of IDV and ATV in plasma were measured by LC-MS/MS.Pharmacokinetics parameters of ddI,d4T,NVP,ATV and IDV in rats and beagles were calculated by statistical moment method.The data were analyzed by Microsoft office Excel 2003 and origin 7.5.Results:①Compared with rats control group,the AUC0-tand Cmaxof NVP in TC group had no obviously change,while the AUC0-tand Cmaxof ddI decreased,but the difference didn't show any statistical significance,and the AUC0-tand Cmaxof d4T was significantly decreased (p<0.05).②The AUC0-tand Cmaxof ATV in rats and beagles after p.o.administration of WZ was significantly increased,compared with the corresponding AUC0-tof ATV before WZ (p<0.05).③The AUC0-tand Cmaxof IDV beagles after p.o.administration of WZ was significantly increased too,compared with the corresponding AUC0-tof IDV before WZ (p<0.05).Conclusion:This thesis had revealed that TC had no effct on pharmacokinetics of NVP,WZ could slow down the metabolism speed of ATV and IDV,provided the good foundation for the further study in the future,and indicated the possible pharmacokinetics interactions when clinical co-administration natural drugs with chemical drugs.
