Objective : To prepare poly(butyl cyanoacrylate) ( PBCA) nanoparticles loaded with Vinblastine Sulfate(VBL-PBCA-NP)and investigate its anti-carcinoma effect on C6 rat intracerebral glioma cells. The safety of VBL-PBCA-NP was detected .Methods:①VBL-PBCA-NP was prepared by emulsification polymerization with dextran asAstabilizing agent.Transmission electron microscopy was used to detecte the shape and size of nanoparticles. Drug entrapment efficiency and loading efficiency were detected by UV-Solution.②MTT and growth curve method were used to observe the inhibiting effect of VBL-PBCA-NP and VBL on C6 rat intracerebral glioma cells. The ability of formation of klong in single cell was detected by klong formation experient. HE strain and SEM was used to observe the configuration and microstructure of the rat's C6 intracerebral glioma cells. FCM was used to detect the cell cycles and apoptosis under the VBL-PBCA-NP. All of thede experients were compared with VBL.③MTT used to detect the toxicity of the rat's neuroglia cells. The safety of the VBL-PBCA-NP was detected by hemolysis experients and acute toxicity of baica lin injection.Result:①The mean diameter of VLB-PBCA-NP was about 74.4nm, and drug entrapment efficiency and loading efficiency was 74.47% and 18.62%.②Compared with VBL, Growth-inhibiting of VBL-PBCA-NP enhances significantly on C6 rat intracerebral glioma cells (P<0.05) in the area of 0.5~5000μg/L .Result of FCM shows that: C6 rat intracerebral glioma cells apoptosis under VBL-PBCA-NP was significantly higher than that under VBL, main cells of which stay at G2.Simultaneitly, the growth-inhibit ability of neuralstemcells was significantly higher than that under VBL. HE strain and SEM showed that: VBL-PBCA-NP and VBL can induce the apoptosis of the rat's C6 intracerebral glioma cells.③The half-death of theVBL-PBCA-NP is 26.377mg/Kg, higher than VBL (22.7 mg/Kg).VBL-PBCA-NP can reduce the toxicity to rat's neuroglia cells by MTT.Conclusion:VBL-PBCA-NP prepared can accord with the standard of nanoparticles .It can significantly restrain the growth of C6 rat intracerebral glioma cells and has the smaller toxicity to mices.
