Brucea javanica(L.)Merr is a traditional Chinese medicine, which distributed in some provinces, such as Hainan, Guangdong, Guangxi and Yunnan in China. It has the function of curing heat and detoxification, antimalarial and dysentery, as well as corroding wart.According to the recording of"The Bencao Gangmu"(The oldest Chinese Catalog of Herbs), the B. javanica can be used for treatment of dysentery, tenesmus and hemorrhoids. The chief constituents are quassinoids, with anti-tumor, anti-malarial and anti-amoebic activity. The brusalol,bruceine D and bruceoside A are the main anti-cancer active components. However, all published research papers are more concerned about the brusalol so that it has been studied in detail, but there are a few research works about the bruceine D, therefore, it is necessary to have more studies about the bruceine D.In term of"the traditional Chinese medicine dictionaries"recording, the bruceine D has the special function of antimalarial, and"the handbook of chemical compositions in the Chinese traditional medicine plants"said that the bruceine D is a poison drug to resisting the ameba dysentery. This thesis is focused on the extraction and separation of bruceine D, the alcohol extracting compound from the B. javanica, and the cell experiments have been carried out to find its anti-tumour activity. In our cell experiments, two kinds of human cancer cells have been tested, namely K562 and MCF-7 cells.About the K562 cell experiment, three kinds of concentrations, 5×10-6 mol/L, 5×10-5mol/L and 5×10-4mol/L, of bruceine D have been used, and the inhibitory rate is 20.04%, 22.96% and 27.76%, respectively. The experiment result shows that the bruceine D has only weakly activity for K562 cell.As to the MCF-7 cell experiment, three kinds of concentrations, 5×10-6 mol/L, 5×10-5mol/L and 5×10-4mol/L, of bruceine D have also been used, and the inhibitory rate is 57.78%,87.75% amd 93.24%, respectively. The experiment result shows that the bruceine D has very strong activity for MCF-7 cell.Our research results have shown that the bruceine D has a strongly selective activity for different cancer cells, and it could be expected to be an anti-cancer new drug in the future.
