Effect Of CAHB On Apoptosis Of MUTZ-1 Cells And Its Mechanism

Myelodysplastic syndrome(MDS)is a group of highly heterogeneous disorders characterized by myeloid cell dysplasia and ineffective hematopiesis.It occurs at a high incidence in the elderly population.The molecular pathogenesis of the disease is poorly understand and its prognosis is poor.The response rate for most drugs remains lower level.CAHB,a new agent for MDS treatment,is a differentiation induce and shows a good response in phaseâ… clinical trial.This paper makes a research to explore the effect of CAHB on the apoptosis of MUTZ-1 cells and its mechanism.ObjectiveTo explore the effect of CAHB on the apoptosis of MUTZ-1 cells and its mechanism in vitro.MethodsIn order to analyze the inhibitory effect of CAHB on the growth of human MDS-RAEB cell line MUTZ-1 and to explore the possible cellular and molecular mechanism,the changes of morphologic features of MUTZ-1 ceils are investigated by cytomorphology,we use MTT assay to measure the growth inhibition ratio,use Flow cytometry to detect the changes of cell cycle distribution,use RT-PCR methods to detect the mRNA expression of Survivin,Bcl-2,P53 gene.ResultsMTT assay showed treatment with CAHB remarkably inhibited the growth of MUTZ-1 cells in a dose-dependent and time-dependent manners.The typical apoptotic morphologic features appeared in cells treated with CAHB.Flow cytometry proved MUTZ-1 cells had a dose-dependent apoptosis after cultured with CAHB for 24 h,CAHB treatment decreased S phase cells,increased G₂/M phase cells,but did not change G₀/G₁ phase cells,which indicated arrest in G₂ phase.RT-PCR showed that P53 mRNA was up-regulated and Survivin,Bcl-2 mRNA down-regulated in MUTZ-1 cells.ConclusionCAHB can inhibit MUTZ-1 cells growth,and can induce MUTZ-1 cells apoptosis;Up-regulation of P53 and down-regulation of Bcl-2,Survivin in MUTZ-1 cells may be one of the mechanisms that CAHB induces apoptosis of cells.
