Study On Chemosensitivity Of Peripheral Blood Lymphocytes And Granulocytes Of Tumorpatients In Vitro

Object:To study chemosensitivity of anticancer drugs in the peripheral blood nucleated cells(lymphocytes and granulocytes)of tumor patients,and to evaluate the correlation or different sensitivities between the anticancer drugs,to try to screen better chemotherapeutic drugs or combined chemotherapy drugs.Methods:The sensitivity or resistance of 15 kinds of commonly used chemotherapeutic drugs and they combined chemotherapy in peripheral blood lymphocytes and granulocytes of 129 cases with tumor in vitro by the classic MTT colorimetric assay.Results:The individual chemosensitivity of NSCLC,gastric cancer, colorectal cancer and nasopharyngeal carcinoma were compara tively different. About non-small-cell lung cancer(NSCLC):the chemosensitivity of combined drugs was apparently superior to single drugs,P＜0.05.The sequence for the average inhibition ratio of drugs was TAX＞CBP+TAX＞DDP+NVB＞DDP＞NVB＞CBP.About gastric cancer:the chemosensitivity of combined drugs was apparently superior to single drugs,P＜0.05.The sequence for the average inhibition ratio of drugs in lymphocytes was DOC+5-Fu+DDP＞DOC＞EPI+5-Fu+DDP＞5-Fu＞DDP＞EPI.The sequence for the average inhibition ratio of drugs in granulocytes was DOC+5-Fu+DDP＞EPI+5-Fu+DDP＞DOC＞5-Fu＞DDP＞EPI.About colorectal cancer:the chemosensitivity of combined drugs was apparently superior to single drugs,P＜0.05.The sequence for the average inhibition ratio of drugs in lymphocytes was CPT-11+5-Fu+CF＞ OXA＞OXA+5-Fu+CF＞CPT-11＞5-Fu＞CF.The sequence for the average inhibition ratio of drugs in granulocytes was OXA＞CPT-11+5-Fu+CF＞OXA+ 5-Fu+CF＞5-Fu＞CPT-11＞CF.About nasopharyngeal carcinoma:the chemosensitivity of combined drugs was apparently superior to single drugs, P＜0.05.The sequence for the average inhibition ratio of drugs was GEM+DDP＞5-Fu＞DDP +5-Fu＞GEM＞DDP.Conclusions:The tumour chemosensitivity test in vitro gave some prediction and guidances for the clinic chemotherapy,and it could discover the drug resisting cases.The combined chemotherapy should be selected for NSCLC,gastric cancer and colorectal cancer patients.And adjuvant chemotherapy should be selected for nasopharyngeal carcinoma patients.
