Objective:By comparing the genes of nasopharyngeal carcinoma metastasis and non-distant metastasis,the differential gene expression profiles were screened out by using cDNA Microarray and the mechanism of NPC metastasis was been investigated.Methods:The establishment of nasopharyngeal carcinoma metastasis Group A is the experimental group,the NPC Group with non-metastasis as the group B is the control group,nasopharyngeal benign for the group C.Each sample's RNA was extracted and the quality of them was identificated.Each equivalent RNA from each group of samples was mixed a sample pool, using the reverse transcriptase method mades cDNA chain.Two probes,labeled by Cy3 or Cy5 fluorescent dyes,hybridized with a human SBC microarray contained 16 KcDNA.Differential gene expression profiles of them were ananlyzed by biostatistics and bioinformatics,then check it by real-time fluorescent quantitative PCR.Results:Three groups of analysable data revealed the fact,Totally 134 two-fold differentially expressed genes were screened out by SBC human genome cDNA microarrays between NPC metastasis and it's non-metastasis, including 107 up-regulated genes and 27 down-regulated genes.Genes involved in cell adhesion genes,extracellular matrix proteins,cell cycle genes,cell growth and proliferation genes,apoptosis genes,transcription factors and regulations,etc,with the verification of Real-time quantitative PCR,PTK2 and other six genes' expressions were consistent with the gene chip's.Conclusion:Nasopharyngeal carcinoma metastasis-associated gene expression profiles were Screened out by gene chips,and providing a theoretical basis to understand the mechanism of NPC metastasis and judge clinical prognosis.
