ObjectivePulmonary fibrosis(PF)is a kind of diffuse interstitial lung disease which caused by many causes and has similar pathogenesis,Its pathogenic characterize is which alveolar structure destruction and diffuse alveolitise in the early,then cause over aggradation of ECM and gaudily leading to fibrosis,consequently cause respiratory function destruction.Most of the patients died from respiratory failure from progressive pulmonary fibrosis.Its pathogenesis is not thoroughly understand till now,so recently the incidence rate and the death rate of pulmonary fibrosis has trend of increasing.The effects of traditional medicine of Glucocorticoid and cell toxicant is dissatisfactory to treat pulmonary fibrosis,so at present,the urgent question affronting the medical science field is to seek effective medicine for pulmonary fibrosis.The research show that curcumin possess widespread pharmacological action in recently years,for example anti-inflammatory,anti-oxidation,anti-ischemic, anticancer,inhibiting hepatic fibrosis.To discuss the preventing and treating effect of curcumin on pulmonary fibrosis,this experiment was adopted MRC-5 induced by transforming growth factor 1(TGF—β1),to explore the effect and mechanisms of curcumin on PF from molecular level in vitro,which can provides experimental foundation for clinic therapy.MethodsMTT colorimetry was used for assaying proliferation of MRC-5.The proliferation curves of MRC-5 were drawn in a week.After cells were incubated with different concentration of curcumin for 48 hours,the inhibiting effects of curcumin were determined by absorbance at 570nm.the MRC-5 were divided into four groups:normal group,cells cultured with MEM/NEAA;curcumin group normal group,cells added with curcumin;TGF-β1 group,cell cutured with TGF-β1(5ng/ml);TGF-β+Cucurmin group,cell cultured with medium added the TGF-β1+Curcumin.The mRNA expression of SMAD3;SMAD 4;SMAD7 these mRNA were tested by the Real-time PCR and the reverse transcription reaction.ResultsIn the normal group MRC-5 express all of the gene just as mentioned above When its culture medium plused curcumin for 48 hours the mRNA of TGF-βRⅠ, TGF-βRⅡexpress strength.the TGF-β+Cururmin group:when the medium added the curcumin,it is easy to see the inhibition effect.It not only inhibits the cells TGF-βRⅠ,TGF-βRⅡmRNA over expression caused by TGF-β1,but also decreases SMAD3;SMAD 4.on the contrary it increases the mRNA of SMAD7 obviouslyConclusionsCurcumin can inhibit the MRC-5's collagen and fibronection deposition which may have important role in the delaying lung fibrosis forming progress.Its mechanism of action maybe through through the inhibiting TGF-βRⅠ,TGF-βRⅡand SMAD3;SMAD 4;however increasing SMAD7 expression.
