Objective:To set up rat model of methamphetamine dependence,and to research HSP70 and Gα12expression in six related encephalic region of methamphetamine dependence rat by mean of immune histochemistry.To explore the relationship between the expression of HSP70 and defend mechanism when nervous system be damaged by methamphetamine,to explore the relationship between the expression of Gα12and methamphetamine dependence.It would offer a scientific evidence for neurotoxicity damage and dependence mechanism of methamphetamine.Methods: Thirty male Sprague-Dawley rats were randomly assigned into control(n=10,saline) group and two experimental(n=20,methamphetamine)groups.The experimental group was administered methamphetamine(ip)once a day with an dose of 10 mg·kg-1(n=10)each time and 20 mg·kg-1(n=10)each time in day 10.On the time to observe stereotyped behavior and do experiment of CPP so that to establish rat model of methamphetamine dependence.Accordingly the control group was injected with saline of the same volume.At the time point of day 10 after injection termination,rats in both groups were perfused and the brains were removed at the same time.The sections including nucleus cortex of frontal lobe(PFC),Striatu(CPu),Nucleus accumbens septum(NAc),Comu ammonis(CA),Ventral tegrnental area(VTA) and Substantia nigra(SN)were selected and mounted on sliders.The gray mean levels of HSP70 and Gα12in selected area were analyzied within immune histochemistry and the results were compared according to the identical time points. Results:For the gray mean levels of HSP70 and Gα12,significant difference was observed in the experimental group(P<0.001)except PFC,HSP70 and Gα12in the experimental groups had higher gray mean levels except PFC.Conclusion: After withdrawal of methamphetamine dependence exposure,HSP70 and Gα12 expression in related encephalic region of the rats are increase except PFC,which suggests that HSP70 and Gα12expression of related encephalic region in rats were involved in the development of amphetamine dependence and the defend mechanism of nervous system.
