Objective:Ischemia reperfusion injury is main reason what induced acute or chronic renal failure.How to prevent and cure acute or chronic renal failure is clinically badly problem.Nowadays ischemic preconditioning prevent IRI this effectiveness is confirmed on dog or rat's heart or liver study.IRI is a significant non-antigen dependence factor which is delayed renal graft function,and can increase the hazard index morbidity of chronic allograft nephropathy.T.F.Fuller through ischemic preconditioning can improve the rat graft renal functional recovery which undergoing grave IRI.Takada monitored HGF in renal transplant acute rejection had apparently heightened.HGF can suppress nephric tubule cellular necrosis,promote nephric tubule cellular generation,antagonize renal interstitial fibrosis,maintain normal kidney organize and structure,and promote renal function to recover.On past study and research foundation this study set up rat renal different time ischemia and ischemic preconditioning reperfusion injury model,and definite the ischemic preconditioning intervention effective.Through detect serum HGF this study elucidate the effective and significance of HGF in different ischemic time and ischemic preconditioning reperfusion injury.Methods:1.There are five rat groups at random:different ring clamp left kidney pedicle 0min(ten number),30min(ten),60min(ten),90min(ten),under 24 h of absoluteness reperfusion,detect renal functional parameter serum creatinine and serum urea nitrogen to definite the degree with renal functional injury in each groups. 2.Definite the ischemic preconditioning intervention effectiveness in 60min(IP1)or 90min(IP2) groups.IP method fractionate two sets:IP1 group ring clamp left kidney pedicle two minutes then open blood current ten minutes,ring clamp left kidney pedicle three minutes then open blood current ten minutes,ring clamp left kidney pedicle four minutes then open blood current ten minutes,ring clamp left kidney pedicle five minutes then open blood current ten minutes,and ring clamp left kidney pedicle six minutes then open blood current ten minutes before ring clamp left kidney pedicle 60minutes,then undergo 24 h absoluteness reperfusion;IP2 is the same way but different is ring clamp 90minutes.They are contrasted with ischemia 60min and 90min groups,and compared the ischemic preconditioning effectiveness.3.To take left part nephridial tissue do hematoxylin and eosin dyeing.Under common practice pathology checking and under Paller's nephric tubule score that definite the degree of injury in pathology in each groups.4.Detect each groups rats's serous HGF,understand the effectiveness and significance of HGF in different ischemic time or ischemic preconditioning reperfusion injury.Result:1.Undergoing 30 min,60min,90min of ischemia and 24 h of reperfusion groups in Ischemia Reperfusion Injury study modeling,there is no significant in the 30 min group which compared with control group in statistics;there are deviation in each indicatrix of renal function or the pathological scale in the 60mmin or 90min groups which compared with control group;there are deviation in each indicatrix of renal function or the pathological scale in the 90min group compared with the 60min group.2.In ischemic Preconditioning(IP1 group)there are significant distinction each indicatrix of renal function or the pathological scale which compared with 60min group,and no significant distinction in item of renal function compare with control group,but there are deviation in pathological scale compare with control group;In ischemic preconditioning(IP2 group)there are no significant distinction each item of renal function or the pathological scale which compared with 90min group.3.HGF is the most obviously increased undergoing 60 min of ischemia and 24 h of reperfusion which caused severity injury in rat kidney.Next HGF is higher heightened in IP1 group.HGF is not heightened undergoing 90 min of ischemia which caused terrier severity injury and which destroyed generation of HGF,then HGF descend tendency.HGF is most sensitive indicatrix compared with renal functional parameter in renal functional restoration.Conclusion:1.Undergoing 60 min of ischemia and 24 h of reperfusion group is more fit other undertake research in IRI.2.Ischemic preconditioning(IP1 group)can effectively relieve the ischemia reperfusion injury of kidney,but IP2 group cannot relieve.3.HGF is the most obviously increase undergoing 60 min of ischemia and 24 h of reperfusion which caused severity injuries in rat kidney.Next HGF is higher heightened in IP1 group.HGF is not heightened undergoing 90 min of ischemia which caused terrier severity injuries and which destroyed generation of HGF,then HGF descend tendency.
