Objective To investigate the effect of nAChR on LPS-induced secretion of cytokines and activity of microglial cells with the nAChR agonist,nicotine and nAChR antagonist,mecamylamine.Methods Cultured rat microglial cells were divided into 4 groups:blank control (Group C),LPS(Group L),nicotine+LPS(Group N),mecamylamine+LPS(Group M).Cell-free supematants were collected and assayed for TNF-αand IL-1βrelease by ELISA after cells were charged for 4h and 24h.Cultured microglial cells were plated onto sterile glass cover slips,and OX-42 was accessed by immunohistochemistry analysis.Results LPS-induced TNF-αand IL-1βproductions in Group L were significantly higher than those of Group C(P<0.05).The expression of OX-42 in Group L was significantly enhanced than that of Group C(P<0.05);LPS-induced TNF-αand IL-1βproductions in Group N decreased significantly than those of Group L,and the expression of OX-42 was significantly lessened in Group N than Group L(P<0.05); Compared with Group L,LPS-induced TNF-αand IL-1βproductions in Group M changed insignificantly,and the expression of OX-42 was not significantly changed too(P>0.05).Conclusion The result suggested that LPS can activate microglial cells,increase the secretion of TNF-αand IL-1β,and enhance the expression of OX-42.nAChR is involved in the expression of cytokines induced by LPS in microglial Cell.The agonist,nicotine can inhibit expression of pro-inflammatory mediators induced by LPS in microgial cell.
