Objectives: Several studies have suggested that all-trans retinoic acid (atRA) can induce vascular smooth muscle cells apoptosis inhibit restenosis after Balloon Angioplasty vascular injury. The vascular endothelium is an active, dynamic tissue that controls many important functions, including regulation of vascular tone and maintenance of blood circulation, fluidity, coagulation, and inflammatory responses. Injury of vascular after angioplasty can result restenosis. To keep the proliferation restore the function of vascular endothelial cells are important of reducing restenosis. We investigated the effect of atRA on proliferation and nitric oxide(NO)release of endothelial cells, trying to evaluate its value of inhibiting restenosis.Methods: Cultured cells were divided into five groups randomly, one of contral group, the other four groups were 10-9 , 10-8, 10-7 , 10-6mol/l. Effect of ATRA on proliferation in human umbilical endothelial cells (ECV-304)were detected by flow cytometry and MTT at 24h, 48h,72h and NO production were detected.Results:①ATRA promoted proliferation activity of ECV-304 between 10-9 moL/L and 10-7moL/L in a concentration-dependent manner (n =5, P≤0.01).However, in higher concentration(10-6mol/L), ATRA showed slight inhibition effect to ECV-304.②NO production was increased, when endothelial cells were incubated ATRA with more than 10-9nmol/L. However, in higher concentration (10-6mol/L), ATRA showed slight inhibition effect to ECV-304. All the effects of ATRA were dose-dependent and time dependent.Conclution: ATRA can promot the proliferation and nitric oxide production of ECV-304.
