Objective: Our research is designed to find out how the AngⅡinduce the release of superoxide anion and PAI-1 in human umbilical vein endothelial cells? Whether the production of PAI-1 is related with the augment of superoxide anion?Methods: we choose the ECV304 as the objective cells , The experiment was divided into three parts ,one is to test the change of superoxide and PAI-1 production under the stimulation of angiotensinⅡ,another is to obersve whether the inhibitor of AT1 receptor Valsartan can prevent the effect of angiotensinⅡ,the third step is to survey the relationship between the production of superoxide anion and PAI-1 with the intervention of Valsartan,DPI,SOD .Results:1.The impact of angiotensinⅡon the production of superoxide anion(02- ) and PAI-1 in HUVECs。Compared to the controlled AngⅡc an promote the production of 02- and PAI-1,And the effection has a positive relateion with the concentratin of angiotensinⅡfrom 10-8 to 10-5mol/L.The optimization concentration is 10-6mol/L ,and rearch its peak at 24 hours[the concentration of 02- and PAI-1 is (186.03±9.32 )U/L and (141.00±4.82)ng/mL],compared the controlled group[the concentration of 02- and PAI-1 is (62.33±5.85)U/L and(60.67±3.51)ng/mL)] has a significant difference(P<0.05)。2.Though valsartan could not change HUVECs's production of superoxide and PAI-1 , but it can prevent the effection of angiotensinⅡeffectively,and the effect also have a concentration dependent relationship within the range from 10-8mol/L to 10-5mol/L.The most effective concentration of valsartan is 10-6mol/L ,compared to the group which cultivation only with antiotensinⅡ[the concentration of 02- and PAI-1 respective is(163.56±5.50)U/L and (151.56±7.88)ng/mL],valsartan in 10-6mol/L can inhibit the angiotensin induced production of 02- and PAI-1 significantly [the concentration respective is(76.20±2.52)U/L and (78.51±4.32)ng/mL].3.DPI a special inhibitor of NADPH oxidase and SOD a scavenger of superoxide anion both can attenuate AngiotensinⅡinduced superoxide anion significantly ,in the same time they also decreased the production of PAI-1 .Conclusion: 1.AngiotensinⅡcan promote the production of superoxide anion and PAI-1 ,the effection has a positive relateion with the concentratin of angiotensinⅡand the stimulate time.2.Valsartan and DPI can prevent the effection of angiotensinⅡeffectively,So we believe that the effection was performed through angiotensin receptor type 1 to activation NADPH oxidase.3.We found that the release of PAI-1 is related with the production of superoxide anion ,PAI-1 decreased when we inhibited or scavenger the production of superoxide aion .It indicate that we can use antioxidator to prevent the incidence of thrombus.
