Objective : To investigate the expression and significance of pigment epithelium-derived factor in choroidal neovascularization(CNV) induced by argon laser on Brown Norway rat.Methods:There were 36male BN rat, we divide their right eyes into experimental group and left eye for control group. And we performed photocoagulation by argon laser in single eye to induce CNV. Three groups with 12of each were randomly formed. And their eye ball was harvested after 1, 2, and 3 weeks as specimen. We perform pathological examination and immunohistochemistry examination of Fâ…§R:Ag to detect the formation of CNV. And the expression of PEDF in the formation process of CNV was detected by immunohistochemistry and Enzyme Linked Immunosorbent Assay.Results:CNV formation was started at 1 week after photocoagulation, and gradually increased at 3 weeks. The positive staining density of Fâ…§R:Ag in light spot region increased gradually (P<0.05), which indicated that the model was successfully estabished. By immunohistochemistry examination of PEDF in each eye ball specimen, the positive staining density of PEDF decreased gradually (P<0.05), PEDF showed a negative correlation with the positive staining density of Fâ…§R:Ag (r=-0.832,P<0.05). By Enzyme Linked Immunosorbent Assay of each eye ball specimen, the concentration of PEDF in each specimen was detected. All the specimens of protein concentrtion of PEDF were less than the control groups(P<0.05). And as time went by, the protein concentrtion of PEDF in each specimen decreased gradually(P<0.05).Conclusion: The formation of CNV showed a negative correlation with the expression of PEDF, which indicated that the deficiency of PEDF expression maybe a foundamental reason for formation and proliferation of CNV.
