Objective: To investigate the inherent relation between the activity of cholinesterase of brain tissue and the expression of Aβ, ChAT of brain tissue with Alzheimer disease(AD) in rats with type-2 diabetic mellitus(DM).Methods:40 rats were randomly divided into four groups:diabetic model group for 2 weeks and it's control group, diabetic model group for 6 weeks and it's control group, every group had 10 rats.Type-2 diabetic rats were induced by high-fat and high-caloric diet for 12weeks and a very small dose of streptozotocin(STZ) through intravenous injection. 2 weeks and 6 weeks later,to take the blood from abdomen aorta, then measured FBG,FINS in the serum, and then to get the brain, and select the left frontal lobe, temporal lobe and hippocampus for immunohistochemistry of ChAT and Aβ, separating the right side cerebral cortex to measured activity of AchE,BchE.Results:1. Type-2 diabetic rats were successfully induced by high-fat and high-caloric diet and a very small dose of streptozotocin(STZ) through intravenous injection.2.The serum level of FBG,FINS in each DM group was significantly higher than that of the control group(P<0.01).The activity of AchE,BchE of brain tissue in DM group for 6 weeks lower than that of the control group(P<0.01).3.The result of expression of Aβof immunohistochemistry revealed that the grey scale in hippocampus,frontal lobe and temporal lobe of each model group was lower than that of the control group,which suggested that the deposition of Aβwas more than that of the control group(P<0.01). The result of expression of ChAT of immunohistochemistry revealed that the grey scale in hippocampus,frontal lobe and temporal lobe of each model group was higher than that of the control group,which suggested that the amount of ChAT was lower than that of the control group(P<0.01=.Conclusion:The decreasing of cholinesterase activity of brain tissue, increasing of expression of Aβand decreasing of expression of ChAT in type-2 diabetic rat are similar to some characteristics of AD to some extent,which suggested that type-2 DM may be related to the development of AD and raise the risk of occurrence of AD.
