Objective:To explore the protective effects of the liver preservation solution which contains fluorocarbon on liver during the process of preserving liver by machine perfusion.Methods:Sixty male Wistar rats were randomly divided into 3 groups:control group 1 (group A,n=20),control group 2(group B,n=20) and fluorocarbon group(group C, n=20).Donor liver is simple cold storage(SCS) in group A.In Group B and C,liver is preserved respectively by perfusing HCA solution and self-made solution which contains fluorocarbon. After the model of cold machine perfusion(CMP) established,The changes of alanine aminotransferase(ALT),tumor necrosis factor(TNF-a) and hyaluronic acid(HA) absorption in reperfusion solution were tested after preservation 12h.Sinusoidal endothelial cell(SEC) were observed under light microscopy and electron microscopy by using isolace perfused rat liver model.It shows that the self-made solution which contains fluorocarbon is better to preserve liver.Results:ALT and TNF-a were both lower in group B and C than in group A,HA absorption was higher.They were lower or higher in group C than in group B.There was significant difference between A and B,B and C,A and C(P<0.05).Along with the time of preservation going on in control group 1,the histopathologic changes gradually aggravated,the changes included degeneration and necrosis of liver,SEC was destroyed.The histopathologic changes of liver in control group 2 and experimental group was lightener than in control group 1.The change in experimental group was lightenest.Conclusion:Self-made solution which contains fluorocarbon has better effects than SCS and preserving by perfusing HCA solution.CMP can lessen SEC injury and reperfusion injury as well as prolong preservation time effectively.
