Objective:To determine the expression level and variability of BMPRs,BMP-2 and Noggin in the human osteoblastic cell line hFOB 1.19 and the human osteosarcoma cell line MG-63.Observe the effect of different dosages exogenous Noggin proteins on the proliferation of the osteosarcoma cells in vitro.Investigate the possible mechanisms of the inhibition effect of noggin on the proliferation of osteosarcoma cells.Provide the basis for the clinical application of Noggin in osteosarcoma therapy.Methods:Determine the expression level of BMP-2,BMPRs(BMPR-IA,BMPR-IB and BMPR-Ⅱ) and Noggin mRNA in the human osteoblastic cell line hFOB 1.19 and the human osteosarcoma cell line MG-63 by RT-PCR method.then observe the effect on the proliferation and morphologic change of the MG-63 cells after given different dosages of exogenous recombinant Noggin protein(1μg/ml,10μg/ml and 20μg/ml) by inverted phase contrast microscope and MTT method.Results:BMPR-IA and BMPR-ⅡmRNAs were detected in both hFOB 1.19 and MG-63 cells.The expression level of BMPR-IA and BMPR-ⅡmRNAs was higher in human osteosarcoma cells than that in human osteoblastic cells.But BMPR-IB mRNA was not detected in both cell lines.Endogenous BMP-2 and Noggin mRNAs were detected in both cell lines.The expression level of BMP-2 mRNA was significant higher in MG-63 cells than that in hFOB 1.19c cells(P<0.05).The variability of Noggin mRNA was not significant(P>0.05).and after given the exogenous Noggin protein,the proliferation of the osteosarcoma cells was inhibited in a dose and time dependent fashion.Conclusion:There're different expression patterns of BMPR,BMP-2 and Noggin between human osteosarcoma cells and osteoblastic cells.The proliferation of the osteosarcoma cells can be significantly inhibited by exogenous noggin protein in vitro and the proliferation of the osteosarcoma cells was inhibited in a dose and time dependent fashion.Thus,Noggin could be one of the possible targets for the therapy of the human osteosarcoma.
