| Objective: To investigate the trends and influential factors of striatal dopamine and glutamate in cell implantation for Parkinsonian rats, and to compare the therapeutic efficacy of embryo ventral mesencephalon (VM) cell with cortico-neural stem cell (NSC).Methods: The striatal dopamine and glutamate were detected by microdialysis combined with high performance liquid chromatograph (HPLC); and the changes of animal ethology and histopathology were surveied and evaluated after cell implantation.Results:①The basal level of striatal dopamine and glutamate in normal rats is about 52.43nmol/L and 2.33μmol/L respectively.②The method to exchange probe shows smaller influence to target neurotransmitter than the method to remain probe in intracerebral microdialysis, and more profit to reclaim probe effectively.③Neither narcotization nor transplant operation has influence to the striatal dopamine and glutamate in normal rats; but Ketamine may causes a transient increase for the striatal glutamate in extracelluar fluid.④Establish the Parkinsonian(PD) rats successfully by giving unilateral stereotaxic injections of small dose 6-hydroxydopamine(6-OHDA) into the substantia nigra pars compacta(SNc) and the medial forebrain bundle(MFB). Moreover, the basal level of striatal dopamine in PD rats is about 7.99nmol/L, which is lower than normal rats'significantly; and the basal level of striatal glutamate, about 10.22μmol/L, is higher than normal rats'.⑤4 weeks after implanted VM cells into striatum of PD rats, the striatal dopamine is no longer decrease, simultaneously the apomorphine(APO)-induced rotational behavior has a significant improvement. Immunohistochemical study 8 weeks after implantation shows VM cells could survive and migrate, even differentiate into tyrosine hydroxylase(TH)-positive neurons.⑥There was no improvement for the striatal dopamine and the APO-induced rotation after implanted NSC cells into striatum of PD rats. Furthermore, the immunohistochemical study later stage shows NSC cells could survive and migrate, but not differentiate.Conclusion:①The method to exchange probe shows smaller influence to target neurotransmitter than the method to remain probe in intracerebral microdialysis, and more profit to reclaim probe effectively.②Neither narcotization nor transplant operation has influence to the striatal dopamine and glutamate in normal rats.③The embryo ventral mesencephalon cell(VM) is better than the cortico-neural stem cell(NSC) for the treatment of Parkinson's disease. |
