Objective:To evaluate the effects of intraperitoneal injection of CpG oligodeoxynucleotide(ODN) on the expression of Th2-specific transcrip- tion factor GATA-3 in lung tissue and allergic airway inflammation in asthmatic mice;and to investigate the mechanism of CpG ODN in the prevention and treatment of bronchial asthma.Methods:Thirty-two BALB/c mice were randomly divided into four groups: normal control group,asthma model group,CpG ODN treatment group and control ODN treatment group,eight mice per group.The mouse model of asthma was reproducted by immunization with intraperitoneally injected and inhaled chicken ovalbumin.CpG ODN was injected by peritoneal cavity one hour before challenged. The total cell count,eosinophil percentage(EOS%) were checked and the concentration of IL-4,IFN-γwere detected by ELISA in bronchoalveolar lavage fluid(BALF).The airway inflammation was observed by hematoxylin and eosin staining.The expression of GATA-3 in lung tissue was detected by immunohistochemical staining and RT-PCR.Results: Compared with asthma model group, the airway inflammation was remarkably relieved,the total cell count,EOS%,concentration of IL-4 in BALF and the expression of GATA-3 in lung tissue decreased, while the concentration of INF-γin BALF increased in CpG ODN treatment group (P<0.01).There was no significant difference between contral ODN treatment group and asthma model group(P>0.05).Conclusion: Intraperitoneal injection of CpG ODN can remarkably relieve the allergic airway inflammation in asthmatic mice,its mechanism may be related to inhibiting the expression of GATA-3 which induce the production of Th2-type cytokines and reduce the production of Th1-type cytokine,and regulating the balance of Th1/Th2.
