| Objective: To investigate the relation between TFF3 and TR3 expression and the carcinogenesis, progress and metastasis of gastric carcinoma, and to explore the related mechanisms.Methods: (1) The total RNA were extracted from SGC-7901 and GES cells, then cDNA were synthesized respectively, the primer of TFF3 was designed and synthesized, RT-PCR was used to detect the expression of TFF3 mRNA in both kinds of cells. (2) Western-blotting was used to detect the expression of TFF3 protein in SGC-7901 cells, GES cells, gastric carcinoma and normal gastric mucosa respectively. (3) Immunohistochemistry was used to detect the expression of TFF3 protein in SGC-7901 cells and paraffin section of gastric carcinoma. (4) The expression of TFF3 protein and TR3 protein in 99 case of gastric carcinoma and 30 case of normal gastric mucosa were detected by Tissue microarray. The relation between TFF3 and TR3 expression and the metastasis and clinicopathological staging of gastric carcinoma was analyzed through statistical method, and the correlation between TFF3 and TR3 was analyzed in further.Results: (1) TFF3 mRNA was highly expessed in SGC-7901 cells and gastric carcinoma, however, it could not be deteced in GES cells and normal gastric mucosa. (2) TFF3 protein could not be deteced in SGC-7901 cells, GES cells, gastric carcinoma and normal gastric mucosa, the negative results probably due to low molecular weight of TFF3, small percentage of wild type TFF3 in total protein, or some steps in western-blotting method needed to be improved. (3) TFF3 protein was highly expressed in SGC-7901 cells, and was higher in gastric carcinoma compared with normal gastric mucosa; furthermore, the level of TFF3 expression was in gastric carcinoma. (4) In gastric carcinoma, the expression of TFF3 protein and TR3 protein were higher than that in normal gastric mucosa (77.8 %vs 26.7 %, 68.7 % vs 30 % , P <0.05). The expression of TFF3 protein and TR3 protein were related to the metastasis and clinicopathological staging of gastric carcinoma. Spearman analysis showed that TFF3 has a positive correlation with TR3 in different clinicopathological staging of gastric carcinoma.Conclusion: (1) TFF3 mRNA was highly expressed in SGC-7901 cells, while it was not expressed in GES cells. (2) Western-blotting method should be improved to detect the TFF3 protein. (3) The expression of TFF3 protein was higher in gastric carcinoma, and the level was correlated with cellular differentiation. TFF3 protien was expressed higher in well-differentiated gastric carcinoma than that in poorly differentiated ones. (4) The expressions of TFF3 and TR3 in gastric carcinoma were closely correlated with pathological stage. (5) TFF3 had a positive correlation with TR3 in gastric carcinoma. |
