Purpose:To investigate the expression and diagnostic value ofβ-catenin,the key of the Wnt signal pathway,and bcl-2,cyclin D1,c-myc,in solitary fibrous tumor(SFT)Methods:1.Materials:22 cases of SFT from Department of Pathology,the First Affiliated Hospital of Nanjing Medical University were collected.15 cases were benign and 7 cases were intermediate ones.2.In situ hybridization(ISH)ofβ-catenin oligonucleotide probe was performed on 22 cases of SFT.3.Immunohistochemical markers,includingβ-catenin,bcl-2, cyclin D1 and c-myc,were used in 22 cases of SFT by the SP immunohistochemical staining method.Results:1.Positive staining ofβ-catenin was seen in 14 of 22(63.64% %)cases of SFT by ISH.In benign and intermediate groups, the positive staining rates were 53.33%(8/15)and 85.71%(6/7) respectively,which were of no significant difference between 2 groups(p>0.05).2.Immunohistochemical positive staining ofβ-catenin was seen in 16 of 22(72.73%)cases of SFT.The immunohistochemica -1 positive rates ofβ-catenin,bcl-2,cyclin D1 and c-myc were 72.73%(16/22),81.82%(18/22),63.64%(14/22),18.18% (4/22)respectively,none of which were of significant difference between 2 groups(p>0.05).The strong positive staining rate ofβ-catenin were 31.82%(7/22),and in benign and intermediate groups,it were 13.33%(2/15)and 71.73%(5/7)respectively.The strong positive staining rates between 2 groups were of significant difference(p=0.01).3.The positive staining rates ofβ- catenin between ISH and immuno -histochemistry were of positive correlation(r_s=0.60,p=0.01).Imm -unohistochemically,the positive staining rates betweenβ- catenin and bcl-2,betweenβ- catenin and cyclin D1,between bcl-2 and cycli -n D1 were of positive correlation respectively(r_s=0.23,p=0.04; r_s=0.40,p=0.04;r_s=0.47,p=0.01;respectively).However,the positive expression pattern between c-myc andβ- catenin,bcl-2 and cyclin D1 were of no correlation respectively(r_s=0.14,p=0.25; r_s=0.028,p=0.42;r_s=0.18,p=0.14;respectively).Conclusions:1.The nuclear expression ofβ-catenin(31.82%)indicated that the Wnt signal pathway is activated in the process of the development of SFT.2.The strong positive immunohistochemical staining pattern ofβ-catenin may be an useful marker in predicting the prognosis of SFT.
