With the close relation of aging and the prolonging of the age , the morbidity of Alzheimer's disease is greatly increasing .It is the forth cause of death in old people's disease.The onset and progress of patient's condition is influenced by many factors , the pathogeny is not very clear by now .Some studies suggest that it is effective way of pharmacotherapy to block the expression of iNOS of gliocyte and inhibit the activity of iNOS . Polygona-polysaccharose play an important role in pharmacological , such as anti-free-radical injure , anti-senium and anti-inflammatory and so on . Therefore, mice which had been transferred genes of APP were studied in this experiment. The aim of the present study is to investigate the effects of polygona-polysaccharose on the spatial learning and memory of APP transgenic mice, the influence of polygona-polysaccharose on biochemical indexes and hippocampus structure in APP transgenic mice, the expression of neural NOS (nNOS) and the expression of inducible NOS( iNOS )in APP transgenic mice and the influence of polygona- polysaccharose on them.The purpose is to explore the effects and functions of polygona-polysaccharose on APP transgenic mice, and try to find a new way of preventing and treating experimental AD.Objective: To investigate the effect of polygona-polysaccharose on APP transgenic mice, and to study the protective and therapeutic function of polygona-polysaccharose on APP transgenic mice.Method: Inject different concentration of polygona-polysaccharose solution into the mices' stomach continuously for 45 days. The Morris water maze was adopted to study the ability of the mices' initiative learning and memory; light microscope was used to observe morphology; the expression of nNOS and iNOS was detected by immunochemistry tissue staining method. Eventually, the effect of polygona-polysaccharose on the above-mentioned index was investigated.Result: The experimental result shows that polygona-polysaccharose has the following effects:1. Ethology : compared with the treat group , the delitescence of the Blank group prolonged obviously, the frequency of finding the platform decreased, and the frequency of mistake increased (p<0.01). Within the treat group, the group with high concentration polygona-polysaccharose increased in the frequency of finding the platform, shortened in the delitescence, and decreased in the frequency of mistake (p<0.05).2. Expression of nNOS and iNOS: compared with the Blank group, the energy of nNOS in the treat group was heightened(p<0.01); and the expression of iNOS was degraded (p<0.01). Within the treat group, there is significance difference. (p<0.01).3. Morphology change: the amount of neuron of the blank was less than that of the treat group. Cell Apoptosis phenomenon could be seen, for example, cell nucleus of some neuron showed pyknosis condition, caryotheca slightly thickened and aberration lipofuscin increased and so on. Polygona-polysaccharose treat group had more neurons, the changes of cell were not obvious, cell had normal structure.Conclusion: Polygona-polysaccharose can adjust the expression of nNOS and iNOS and improve the ability of learning and memory of APP transgenic mice. T. Polygona-polysaccharose is an effective prescription for the prevention and therapy of experimental AD.
