Effect of different hemorrhage to Myocardial Emzyme and Expression of Fas in the rats under the controlled hypotensionObjective:To observe the influences of different hemorrhage to myocardial emzyme and expression of fas gene in the rats under the controlled hypotention(CH)induced by sodium nitroprusside(SNP)and esmolol(ESM),to explore the rats' tolerance to different hemorrhage under the controlled hypotension.Method:48 Sprague-Dawley rats weighing 260,350g,are randomly divided into 8 groups(n=6):group C1(hemorrhage of 10%),group C2(hemorrhage of 20%),group C3(hemorrhage of 30%),group C4(hemorrhage of 40%),group CH1(controlled hypotention+ hemorrhage of 10%),group CH2(controlled hypotention+hemorrhage of 20%),group CH3(controlled hypotention+hemorrhage of 30%),group CH4(controlled hypotention+hemorrhage of 10%),The mean arterial pressure(MAP)of groups CH is to be keep 50-55mmhg with sodium nitroprusside(SNP)and esmolol(ESM)for ten minutes,then let blood from the femoral。The groups C are continus infused 0.9% physiological saline solution(NS),then let blood from the femoral.The MAP of all the groups is raised a hour later.Recording vital sign of rats and doing blood gas analsy.Having a laboratory test of myocardial emzyme and detecting the myocardial cell apoptosis and protein expression of fas gene)with TUNEL。and immunohisto chemical staining respectively after 24 hours.The survival rate of the rats are also recorded.Result:1.The level of the LDH and CK in the group CH3 is the highest than other groups(P<0.05).Because of the high death rate,the group C4 and CH4 are excluded.2.The expression of fas genes in the group CH3 is the highest than other groups(P<0.05).3.The expression of the myocardial cell apoptosis in the group CH3 is the highest than other groups(P<0.05).Conclusions:1.The heart of the rats can tolerate the hemorrhage of 10%and hemorrhage of 20%without significant change of myocardial emzyme and expression of fas gene under the controlled hypotensiono When the hemorrhage is upto 30%,the myocardial emzyme and expression of fas gene is increasd significantly.In this case,the myocardial cell is damnified and lead to the apoptosis.2.The rats cann't tolerate the hemorrhage of 40%whether controlled hypotension or not,it's mortality is high。...
