Objective:To study the effect of suppressive oligodeoxynucleotides(Sup ODN)on the Th1 differentiation of CD4+ T splenetic lymphocytes of mice.Methods:The splenetic lymphocytes of BALB/c mice were separated and then CD4+ cells were purified with immune magnetic CD4+ microbeads(positive selection).The purification was examined by FACS.The co-culture of CD4+ cells,anti-CD3ε,anti-CD28,IL-12 and Sup ODN or control oligodeoxynucleotides(Con ODN)was performed for 72 hours.The IFN-γand IL-4 in supernatant were detected using ELISA.The expression of CD4+ cells T-bet mRNA was tested by RT-PCR.Results:Sup ODN significantly respectively inhibited INF-γrelease and increased IL-4 production(P<0.01).T-bet mRNA of CD4+ lymphocytes was remarkably inhibited by Sup ODN as well(P<0.01).Conclusion:In present of pro-Th1-cytokines,Sup ODN may have effects on differentiation of CD4+ T lymphocytes in vitro.Sup ODN can differentiate CD4+ T cells to Th1,also it may suppress the lymphocytes into Th2.
