Objective To study the effect of the frequency of abnormitic sperm morphology, spermatozoa acrosin activities and the amounts of WGA to fertilization rate in vitro. To determine the predictive value of those to fertilization rate of IVF-ET.And help to choose a best protocol before ART.Methods All sperm were stained by diff-quik and analysed according to the WHO strict standard,all patients were divided into group 1(normal morphology≥5%)and group 2(normal morphology<5%).Patients in group 1 and 2 underwent IVF or intracytoplasmic sperm injection(ICSI).Then,the fertilization rates,cleavage rates, good embryo rates and clinical pregnancy rates among each group are statistics.And 35 patients who underwent IVF were divided into two groups,then statistics of morphological sperm,human spermatozoa acrosin activities,the quantities of WGAR on spermatozoa,and the fertilization rate of IVF-ET were made.Results1 During IVF cycles,there was a significant difference between group 1 and 2 on fertilization rates(92.41%vs71.43%,P<0.01).Although cleavage rates,good embryo rates of group 1 were higher than group 2 there was no difference(95.35%vs96.43%,62.57%vs51.85%,P>0.05).2 In ICSI cycles,there was no significant difference between group 1 and 2 on fertilization rates(83.59%vs76.03%),cleavage rates(89.32%vs87.78%),and good embryo rates(63.04%vs51.90%)(P>0.05).3 In group 2(normal morphology<5%),fertilization rates(71.43%vs76.03%), cleavage rates(83.08%vs87.78%),good embryo rates(51.85%%vs51.90%)in ICSI group were higher than those in IVF group,especially cleavage rates were significantly higher(P<0.05).Special morphological sperm such as small size of acrosome sperm and non-acrosome sperm with ICSI protocol,we can obtain better fertilization rates,developmental rates of embryo and clinical pregnancy rates.4 The percent of abnormal morphology head sperm is 77.48%.The head defects is the primacy defect of spem.Significant difference was present in rate of head defect (74.71%vs67.88%)and quantity of abnormal acrosome(92.57vs69.04)between higher fertilization rate group and lower group(71.56vs49.71,P<0.05).The rate of acrosome intactness of higher fertilization rate group is higher than lower group (78.71%vs90.27%,P<0.01).5 Significant difference was present in acrosin activity between higher fertilization rate group and lower group(71.56vs49.71,P<0.05).6 There are positive correlations between acrosin activity and the number of sperm with normal morphology(P<0.05).7 The quantities of spermatozoa surface WGA-Receptor showed significant difference between higher fertilization rate group and lower group(P<0.01).8 By using multiple regression analyses,55.7%of fertilization rates in vitro is determined by changes of WGA-Receptor.ConclusionsSperm morphology,acrosin activity and WGA-receptor are important parameters to estimate the function of sperm.All of those three are helpful to diagnose and the male infertility and choose the treatment of ART.
