Objective:to observe the effect of photodynamic therapy(PDT)on human lung cancer cell line A549 in vitro,and to select the optimization parameters of concentration of photosensitizer and intensity of laser;use the common mathematical quantitative model to simulate the PDT reaction.Material and Methods:The lung cancer cell line A549 cultured in vitro with the culture media F12,and treated with different concentrations of Photosan3 and different dosages of laser,then MTT assay was applied to measure the OD492.The cell survival ratios were calculated and the optimization parameters were selected.To simulate the reaction by the established PDT common mathematical quantitative model.A new type of clear bottom black 96-well plate was used in our study.Results:The cell survival ratios of the clear bottom black 96-well plates were higher than common clear 96-well plates significantly.Photosan3-PDT can obviously inhibit the growth of lung cancer cell line A549;the cell survival ratio was declined followed the increasing of the photosensitizer concentration in a certain range and the trend not in evidence when the concentration under 4mg/l or above 20mg/l.The laser power can also influence the survival ratio from 5mW to 50mW.Our results demonstrate cell death,inhibition of cell growth,and G(0)/G(1)cell cycle arrest during the photosan3 PDT-mediated response.Apoptosis and necrosis were more obvious along with the time prolongation of Photosan3-PDT.Common mathematical model showed an appropriate and reasonable simulation of the typeâ…¡PDT reaction.Conclusions:The clear bottom black 96 plate is appropriate arrangements for the application to PDT cell experiment.The cell survival fraction is declined by the inducement of Photosan3 mediated PDT in the centain range.The inapplicable range affected the down stream obviously.The process and trend of the reaction can be simulated by the common mathematic model correctly.
