ObjectiveTo study the expression of HSPgp96,S100,CD4,CD8 in BTCC and its relationship with the reaction of leukomonocyte in BTCC stroma.Offer new theory of the biotherapy of BTCC.MethodsTo study the expression of gp96,S100,CD4,CD8 in 82 case BTCC tissue and in 4 normal tissues by immunohistoehemieal methods,approch the correlation of expression among gp96,S100,CD4,CD8 in BTCC tissue and the correlation between these index and the reaction of leukomonocyte in BTCC stroma.Results1.The total positive expression rate of Gp96 is 59.3%,The positive expression rate of Gp96 is 25%in normal bladder tissue and 61%in BTCC tissue,the discrepancy of the expression between normal bladder tissue and BTCC tissue is significant(x~2=9.54,P<0.05).The positive expression rate of group of leukomonocyte infiltration and non-leukomonocyte infiltration in stroma is 88.1%,32.5%respectively.The discrepancy has statistical significance(x~2=7.69,P<0.05);the discrepancy between the positive expression rate of the group of leukomonocyte infiltration and non-leukomonocyte infiltration in stroma in each tumor Pathological grades,clinical stages have statistical significance through chi square test and Fisher exact probability test(P<0.05).2.The total positive expression rate of S—100~+Dc is 67.4%,The positive expression rate of Gp96 is 25%in normal bladder tissue and 69.5%in BTCC tissue,the discrepancy of the expression between normal bladder tissue and BTCC tissue is significant(x~2=7.03,P<0.05).The positive expression rate of group of leukomonocyte infiltration and non-leukomonocyte infiltration in BTCC stroma is 90.5%,47.5%respectively.the discrepancy has statistical significance;the discrepancy between the positive expression rate of the group of leukomonocyte infiltration and non-leukomonocyte infiltration in BTCC stroma in each tumor Pathological grades,clinical stages have statistical significance through chi square test and Fisher exact probability test(P<0.05).3.The total positive expression rate of CD4,CD8 is 62.8%,79.1%,The positive expression rate of Gp96 is 0 in normal bladder tissue and 65.9%,68.3%in BTCC tissue,the discrepancy of the expression between normal bladder tissue and BTCC tissue is significant(x~2=8.116,P<0.05),(x~2=6.974,P<0.05).The positive expression rate of group of leukomonocyte infiltration and non-leukomonocyte infiltration in BTCC stroma is 88.1%,42.5%and 88.1%,47.5 %respectively.the discrepancy have statistical significance(x~2=7.112,P<0.05),(x~2=7.034,P<0.05);the discrepancy between the positive expression rate of the group of leukomonocyte infiltration and non-leukomonocyte infiltration in BTCC stroma in each tumor Pathological grades,clinical stages have statistical significance through chi square test and Fisher exact probability test(P<0.05).4.We analyse the correlation between the expression of gp96,S-100,CD4,CD8 and pathological grades,clinical stages of BTCC through Kuskal—Wallis test,the results suggest that the expression of gp96 has positive correlation with pathological grades,clinical stages of BTCC and S-100,CD4,CD8 has negative correlation with pathological grades,clinical stages of BTCC;We analyse the correlation among gp96,S-100,CD4,CD8 through Spearman interclass correlation test,the results suggest that the expression of gp96 has negative correlation with S-100,CD4,CD8, and S-100 has positive correlation with CD4,CD8,the results menthioned above all have statistical significance.ConclusionsThe expression of gp96,S-100,CD4,CD8 in BTCC tissue have correlation with the degree of leukomonocyte infiltration in BTCC stroma,The positive expression rate of group of leukomonocyte infiltration is higher than the group of non-leukomonocyte infiltration in BTCC stroma;The expression of S-100,CD4,CD8 have negative correlation with Pathological grades,clinical stages of BTCC; The expression of gp96 have positive correlation with pathological grades,clinical stages of BTCC,it suggest that the augmentation of level of gp96's expression can be used to be a signal of malignant cell that adjust its antigen characteristics to escape immunologic surveillance,and also suggest that gp96 can be used to be a refer index to judge the degree of differentiation and progression of tumor.
