Objective:To investigate the expression of hBD-1,hBD-2 in the polyp of vocal cord by tissue chip technique and S-P immunohistochemistry, and to further study their role in the pathogenesis of the polyps of vocal cord and the relationship of them.Method:Eighty-five useful tissue chips which include fifty-one polyps of vocal cord,twenty-six vocal nodules and eight normal vocal cords were made by tissue chip technique, S-P immunohistochemistry were used to determine the expression of hBD-1 and hBD-2 in every tissue chip . A mean numbers of positive cells per high power field( 10×40) were counted. All data were analyzed with Krusal-Wallis test and correl-test by SPSS11.5 software.Results:1)The character of our recipient block was exquisite and no splits. Disks were arranged clearly on the slides. After section cutting, the efficient disks were eighty-five of all one hundred and five. Although very few disks had shift or distort, the rate of observation still exceeded 98% after HE staining. The diagnosable epithelium tissues could be found in every tissue chip. The coincidence rate of hBD-2 between the tissue chip and the whole tissue sections were 95.29% (81/85) .There were no significant differences between them (P>0.05).2) hBD-1 was expressed at low levels in all tissue specimens of test group and two control groups. The positive rate of vocal cord polyps was 96.08% (49/51) ,which did not differ significantly with those of vocal cord nodules(96.08% (49/51)) and normal vocal cords ( 87.5% (7/8) )(P>0.05). In the vocal cord polyps, the expressive difference of hBD-1 was insignificant in the male (95.6% (22/23)) and female (96.4% (27/28)) (P>0.05).3) hBD-2 was detected in the specimens of test group , but almost not in the control groups. The positive rate of vocal cord polyps was 100% (51/51 ),which expression level was significantly higher than those of vocal cord nodules (23.08% (6/26)) and normal vocal cords (25% (2/8) )(P<0.05). There were significant differences between vocal cord polyps group and two control groups (P<0.017) with Mann-Whitney U test. But There were no significant differences between vocal cord nodule group and normal vocal cord group (P<0.017). Furthermore, The high positive expressive rate of vocal cord polyps was 62.75%(32/51),which was significantly higher than those of vocal cord nodules (0% (0/26)) and normal vocal cords (0%( 0/8))(P<0.05 ). In the vocal cord polyps, the expressive difference of hBD-2 was insignificant in the male (100%(23/23)) and female(100%(28/28))(P>0.05).4)The expression of hBD-1 and hBD-2 seemed to be positive correlation to vocal cord polyps(P<0.05).Conclusion:1) hBD-1 was expressed in all tissue specimens, at low levels that did not differ significantly (P>0.05). This suggested the expression of hBD-1 was inherent.2) hBD-2 was detected in the specimens of test group, but almost not in the control groups. The positive rate of vocal cord polyps was significantly higher than those of vocal cord nodules and normal vocal cords (P<0.05 ). This suggested the expression of hBD-2 was derivational.3) It is feasible to use tissue microarray technique which is a rapid, economic and accurate method to screen clinical tissue specimens on a large scale.
