Objectives: to evaluate the effect of Rapamycin eye drops on cornea neovascularization of rabbit induced by alkali cauterizationg .Methods: Part 1.to observate the effect of RAPA on HUVMC in vitro.in The inhibitory effect on growth of HUVMC induced by VEGF with different concentrations(0.1~20ng/ml) rapamycin was evaluated by MTT. Part 2.in vivo:Cornea neovascularizationg (CNV)was induced by 1mol/L NaOH in 30 rabbits. The rabbits were then divided into five groups: NO.1 :10mg/L Rapamycin treated group (n=6), NO.2: 20mg/L Rapamycin treated group (n=6), NO.3: 0.25% Dexamethasone treated group (n=6), NO.4: rapamycin menstruum group (n = 6). NO.5: control group (n=6).Each groups have been given the eye drops since the 2 day after alkali injury. the eyes in each groups were examined by microscope at 3,7,14 days.the area of CNV was marked by photography. Immunobistochemistry was used to determine the VEGF and bFGF level .Results: Rapamycin and Dexamethasone eye drops both can inhibit the length and area of rabbit corneal neovascularization at different time,howerer, there was a little difference between the two concentration of rapamycin eye drops.the expression of VEGF and bFGF was suppressed significantly in rapamycin and Dexamethasone eye drops groups,nevertheless the expression of VEGF was markedly increased in the two control groups.Conclusion: the results of this study demonstrate that both of local application rapamycin and Dexamethasone are effective in the inhibition of corneal neovacularization induced by alkaline burn,however,the former appears to more effective than the later.
