| Objective: To invstigate the expression of RhoA protein in two epithelial ovarian cancer cells, HO8910PM and HO8910,and one normal epithelial ovarian cell line HOSEA and its relationship with the invasion and metastasis of epithelial ovarian carcinoma.Methods: Expression of RhoA in two ovarian cancer cell lines, HO8910PM and HO8910,and one normal epithelial ovarian cell line HOSEA was detected by immunohistochemical method. Matrigel invasion assay using three epithelial ovarian cell lines was done in Transwell chambers,so was angiogenic ability assay.Results: (1)The expression levels of RhoA varied in the three cell lines examined. The expression levels of RhoA in HO8910 decreased comparing with HO8910PM but increased comparing with HOSEA(F= 35.73,P<0.01).(2)HO8910PM had the strongest invasiveness,HO8910 was sub-invasive,while HOSEA had non-invasive property(F=105.80,P<0.01).(3)Angiogenic capability of HO8910PM was strongest,HO8910 was weaker,while HOSEA had no angiogenesis.The defference between angiogenic capability of three cell lines was statistically significant(F=24.69,P<0.01). (4)The expression levels of RhoA in ovarian cancer cells was significantly positive correlated with both the invasiveness and angiogenic capability of these cells(r=0.89 or 0.84,P<0.05;r=0.81 or 0.88 ,P<0.05).Conclusion: (1) Among three cell lines, HO8910PM had the highest RhoA expression level,invasion and angiogenic capability, HO8910 was the higher one,HOSEA was the lowest one.(2)The results suggest that expression of RhoA was correlated with both the invasive and angiogenic capabilities of the epithelial ovarian carcinoma cells in vitro. (3)RhoA may serve as an important parameter in the metastasis of epithelial ovarian carcinoma. |
