The Influence To The Gene Express And The Studies Of The Efficacy Enhancing And Toxicity Reducing To Cytoxan By Giant Typhonium Rhiszome On Tumor Bearing Mice

Objective:To study the dead category and mechanism of H₂₂tumor cells by the Giant Typhonium Rhizome for oral use on mice,the efficacy enhancing and the toxicity reducing to cytoxan and the influence on survival quality.Methods:We established the H₂₂tumor-bearing mice model and randomly devided mice into five groups.After treated for ten days,the mice were executed for getting the tumor,thymus and spleen to calculate.We picked up the eyeball for blood to measure the peripheral white blood cells in serum,BUN,Scr,GPT and GOT;got the femoral bones to determine marrow micronuclear rates;observed the morphological alteration of tumor cells by light microscopy and electron microscopy and the impact on the expression of p53 and PCNA in tumor cells by method of immunohistochemistry;observed the situation and survival days to determine the survival quality and life span.Results:â‘ SGTR made the tumor cells apoptosis.â‘¡SGTR can reduce the expression of mt-p53 and PCNA.â‘¢The radio of anti-tumor was 36.80%.The compounds of SGTR and CTX can enhance the anti-tumor effect(q=0.8619).â‘£SGTR did not improve the elevated marrow micronuclear rates and the descent of PWBC after chems.⑤SGTR heightened the loss thymus index induced by tumor.â‘¥SGTR did not reduce the elevated GPT and GOT made by tumor.⑦Both SGTR and tumor did not impact BUN and Scr.⑧SGTR made the weight as increased as the normal mice.⑨SGTR prolonged the life span of tumor-bearing mice,and the rate of life extension was 63.82%.Conclusion:SGTR can regulate the expression of mt-p53 and PCNA in H₂₂tumor cells,so that it make the tumor cells apoptosis to show the anti-tumor function.Combined with CTX,SGTR can promote the effect.SGTR has no toxicity reducing effect on mice bearing H₂₂tumor cells treated with anti-tumor drug like cytoxan.SGTR can prolong the life span,and improve the survival quality.
