Cinnabar was a kind of natural mineral drugs,which had tranquilizing mind and heat-clearing to detoxifying.Mercury sulfide(HgS),the main constituents of Cinnabar,induced mercury poisoning due to long-term,large or improper use of drugs.Mercury could injury Liver,Kidney and the Central nervous system.Mercury poisoning included acute poisoning and chronic poisoning.In clinic,different route of administration,dosage and duration of treatment caused the accumulation of chronic poisoning.Based on the establishment of the determination method of mercury in biological samples,pharmacokinetics,toxicokinetics,tissue distribution and the accumulation of mercury were studied in the body after single and successive bolus oral administration of cinnabar to rats.A preliminary research was explored on the correlation of mercury concentration in the saliva and blood, in the saliva and tissue.The results of this study indicated that the AFS can measure mercury in blood,saliva and tissue samples.The method was simple and rapid, quantification results was reliable in the range of 0.5~20.0μg/L and 0.2~20.0μg/L,respectively.A single compartment model was applied to describe pharmacokinetics of mercury after single large dose of oral administration cinnabar(7.0g/kg)to rats.The tmax,t1/2a,t1/2,were 1.29h,0.28h,6.64h,respectively.After successive bolus oral administration of cinnabar to rats,mercury in blood concentrations achieved a higher level in the fourth day and the tenth day,but there were no significant differences.The concentration of mercury had not increase significantly in heart,lung and liver.In the kidney,the concentration of mercury increased significantly in the tenth day,suggesting that the possibility of accumulation.The results of this study showed that mercury was detected in saliva,the highest concentration of parotid saliva equaled to 7.43%of blood after oral administration 25.5h.The highest concentration of parotid saliva equaled to 7.79%of blood after oral administration 25.5h.The highest concentration of blood equaled to 63.5%,55.7%of liver,kidney after oral administration 26h, respectively.The correlation between SPr(SM)and S/B ratio,SPr(SM)and S/T ratio were good,which suggested that blood and tissue level of mercury could be learned by measuring the concentration of mercury in saliva.The dynamic characteristics of mercury in the blood,the regularity of tissue distribution and excretion in the saliva,and the correlation of mercury level between the saliva" and blood,between the saliva and tissue were researched after single and successive bolus oral administration of cinnabar to rats at the first time.This study provided a practical approach to the determination for the heavy metal poisoning diagnosis,prevention and treatment.
