Expression Of ILK And α-SMA Of DANSHEN Root Intertention In Rats Carbon Tetrachloride-induced Hepatic Fibrosis By Experimental Research

Objective: The aim of the present study is to investigate the efficacy and mechanism of danshen root in a rat hepatic fibrosis model induced by carbon tetrachloride (CCl4).Thus to analyze the expression of integrin-linked kinase(ILK) andα-smooth muscle actin(α-SMA) in the hepatic tissues and further elucidate the mechanism of danshen root in the treatment of hepatic fibrosis.Methods: The model of hepatic fibrosis model was established by administration of CCl4 and was challenged by danshen root simultaneously for 2,4,6,8 weeks respectively.108 rats were randomly divided into three groups by normal drinking and food:12 rat normal control group, 48 rat model control group, 48 rat danshen root group. The liver tissues of rats in all groups (including the control group) were stained by hematoxylin-eosin and were focused on their pathological changes. The expressions of ILK andα-SMA in rat liver tissues were observed with immunohistochemical staining.Results: Showed by immunohistochemical staining , ILK andα-SMA positive cells mainly expressed in hepatic perisinusoidal space of Disse , portal area of fiber proliferation, fiber interval and hepatic perisinusoidal cells near fiber interval. Both were almost expressed in the same position. The expressions of ILK in the normal liver tissues was not significant. Injected by carbon tetrachloride (CCl4), the expressions of ILK andα-SMA were significantly increased in contrast to normal control group, and the intensity of expression was gradually increased in 2,4,6,8 weeks liver tissues(P<0.05). During intervention phase by danshen root in rat hepatic fibrosis, there were few expression of ILK andα-SMA in rat liver tissues. Compared with normal control group , it had mild increase, but no significant difference. While compared to model control group , it showed remarkable decrease( P<0.05). The expression of ILK andα-SMA in all liver tissues of intervention group(danshen root group) were similar and not obviously different(P>0.05). In the hepatic fibrosis model group , the expression of ILK andα-SMA in the tissues were gradually increased , among them were positively correlated(P<0.05) and also positively correlated with the degree of hepatic fibrosis(P<0.05).In the danshen root group , the expression of ILK andα-SMA in the tissues of each group were positively correlated(P<0.05) and also positively correlated with the degree of hepatic fibrosis(P<0.05)。Conclusion: The expressions of ILK andα-SMA in rat hepatic fibrosis were high, which could show that ILK played an important role on rat hepatic fibrosis; Challenged by danshen root in rat hepatic fibrosis, the expressions of ILK andα-SMA were remarkably decreasing, and the result of histology of liver tissue showed the degree of hepatic fibrosis was obviously softened. These showed danshen root might be effective in the treatment of hepatic fibrosis, which displayed danshen root might inhibit the activation of liver stellate cells and the transformation of myofibroblasts. This might provide new view and methods to study the mechanism of protective effect of danshen root in hepatic fibrosis.
