Objective:The study focused on detecting the expression of Estrogen Receptor alph(ERα)and Estrogen Receptor betaβ(ERβ)in bone marrow cells of childhood acute lymphoblastic leukemia(ALL) at the levels of its protein production,and exploring the correlation between the levels of ERα,ERβand ALL.It may help to know the role of ERαand ERβin childhood ALL and may contribute to diagnosis,therapy and prognosis in clinic.Methods: Thirty childhood with ALL were enrolled in newly diagnosed group and the control group included 15 childhood with normal bone marrow.There were remission group and non-remission group after five weeks'united chemotherapies to the childhood with ALL in newly diagnosed group. Fist using an immunocyto- chemical stain technique APAAP with McAb for CD5,CD7,CD10,CD19,CD33,HLA-DR antigens in blast immunophenotyping to the childhood with ALL in newly diagnosed group.The expression of ERαand ERβin all patients'bone marrow cells was detected by S-P immunohistochemical assay .Positive cells were dyed yellow or brown in cytoplasm or nucleus. Positive ERαor ERβwas that the positive cells were >5% bone marrow cells.Results: In 30 childhood patients of the newly diagnosed ALL,the total ERα-positive cases were 17,the positive expression rate was 56.7%(17/30).In 15 conrtol, the total ERα-positive cases were 3, the positive expression rate was 20.0%(3/15).The data were analyzed with chi-square test,χ2=5.45,P<0.05,the former was remarkably significant higher than the latter. In 30 childhood patients of the newly diagnosed ALL, the total ERβ-positive cases were 23,the positive expression rate was 76.7%(23/30). In 15 conrtol, the total ERβ-positive cases were 1, the positive expression rate was 6.7%(1/15). The data were analyzed with chi-square test,χ2=19.69,P<0.05,the former was remarkably significant higher than the latter.By immunocytochemical stain, there were 21 cases of ALL-B and 9 cases of ALL-T in 30 childhood patients of the newly diagnosed ALL. In 21 ALL-B, the total ERα-positive cases were12, the positive expression rate was 57.1%(12/21),In 9 ALL-T, the total ERα-positive cases were 5, the positive expression rate was 55.6%(5/9). The data were analyzed with Fisher exact test, P>0.05,The positive expression of ERαhave no significant difference between ALL-B and ALL-T. In 21 ALL-B, the total ERβ-positive cases were15, the positive expression rate was 71.4%(15/21),In 9 ALL-T, the total ERβ-positive cases were 8, the positive expression rate was 88.9%(8/9),The data were analyzed with Fisher exact test, P>0.05, The positive expression of ERβhave no significant difference between ALL-B and ALL-T.Five weeks'united chemotherapies(VDLP) and development measure were done in 30 childhood patients of the newly diagnosed ALL.,24 childhood patients were remission,in which 14 cases came from the childhood patients with the positive expression of ERαin newly diagnosed group, the rate of the remission was 82.4%(14/17), and in which 10 cases came from the childhood patients with the negative expression of ERαin newly diagnosed group, the rate of the remission was 76.9%(10/13).The data were analyzed with Fisher exact test, P>0.05, the rate of the remission has no significant difference between the positive expression and negative expression of ERα. In 24 childhood patients who were remission,there were 21 cases came from the childhood patients with the positive expression of ERβin newly diagnosed group, the rate of the remission was 91.3%(21/23),and there were 3 cases came from the childhood patients with the negative expression of ERβin newly diagnosed group, the rate of the remission was 42.9%(3/7), The data were analyzed with Fisher exact test, P<0.05, the rate of the remission has remarkably significant difference between the positive expression and negative expression of ERβ.There were 11 cases of positive expression of ERαin the 24 childhood patients who were remission,the rate of the positive expression was 45.8%(11/24), There were 3 cases of the positive expression of ERαin the 6 childhood patients who were non-remission,the rate of the positive expression was 50.0%(3/6). The data were analyzed with Fisher exact test, P>0.05, the rate of the positive expression of ERαhas no significant difference between the remission group and non-remission group. There were 18 cases of the positive expression of ERβin the 24 childhood patients who were remission,the rate of the positive expression was 75.0%(18/24),There were 2 cases of the positive expression of ERβin the 6 childhood patients who were non-remission,the rate of the positive expression was 33.3%(2/6). The data were analyzed with Fisher exact test, P<0.05, the rate of the positive expression of ERβhas remarkably significant difference between the remission group and non-remission group.Conclusions:①The rate of the positive expression of ERα,ERβin newly diagnosed group were higher than that in control group, which means that ERαand ERβmay play a role in the childhood acute lymphoblastic leukemia and may help us explore the mechanism of childhood ALL.②The expression of ERαand ERβmay be no correlation between the type of childhood ALL.③The rate of the positive expression of ERβin remission group was higher than that in non-remission group, which means that the expression of ERβhas a certain correlation to therapy and prognosis of childhood patients of ALL.
