The Study On Expression Of PTEN And PCNA In Gastric MALT Lymphoma And Its Significance

Objective:To study the expression of anti-oncogene PTEN and cell proliferation and nuclear protein PCNA in gastric MALT lymphoma tissues and and compared it with that of chronic tonsillitis organizations CPT and that of normal gastric tissues,to find the relationship between expression deletion of PTEN/PCNA in gastric MALT lymphoma and the pathological features of gastric MALT lymphoma.Method:immunohistochemistry was applied to test PTEN and PCNA protein in 46 cases of gastric MALT lymphoma,46 cases of CPT 46 cases of normal gastric.Results were compared with the expression and organization types,clinical features and clinical stages, and other acts related to biological analysis.Result:Results:1.Expression of PTEN protein:①Positive stain particles of PTEN protein mainly located in the cytoplasm. The positive rate of PTEN in gastric MALT lymphoma group was 47.83%(22/46),The positive rate of PTEN inCPT group was91.3% (42/46),The positive rate of PTEN in the normal mucosa group was95.65%(44/46),The positive rate of PTEN in gastric MALT lymphoma group was significantly lower than CPT organized group and the normal mucosa group(P＜0.05).③expression of MALT lymphoma group was lower than CPT organized group and the normal mucosa group(P＜0.005),Also there was no significant diference between CPT organized group and normal gastric group.④the relationship between TEN protein expression and gastric MALT lymphoma clinical features:The positive rate of PTEN between low-grade group and a high degree of vicious group had a significant difference(P＜0.05);According to clinical pathological stage,The positive rate of PTEN betweenⅠstage Group,Ⅱstage Group,Ⅲstage Groupand N stage Group (85.71%,63.64%,35.00%,25.00%.)Hadasignificant difference (P＜0.05);The positive rates of PTEN decreased with the increase in clinical stage and the increase in the degree of malignancy (P＜0.05).In addition,PTEN protein expression had no relation to the patients'age,sex,tumor size and location.(P＞0.05).2.Expression of PCNA protein:①Positive stain particles of PCNA protein mainly located in the nuclear.②The positive rate of PCNA in gastric MALT lymphoma group was 71.74%(33/46), The positive rate of PCNA in CPT group was was 17.39%(8/46), The positive rate of PCNA in the normal mucosa group was13.04% (6/46).The positive rate of PCNA in gastric MALT lymphoma group was significantly higher than CPT organized group and the normal mucosa group(P＜0.05).③expression of MALT lymphoma group was higher than CPT organized group and the normal mucosa group(P＜0.005).Also there was no significant diference between CPT organized group and normal gastric group.④the relationship between PCNAprotein expression and gastric MALT lymphoma clinical features:The positive rate of PCNA between low-grade group and a high degree of vicious group had a significant difference(P＜0.05);The positive rate of PCNA between the gastric MALT lymphoma not invaded through serous and invaded through serous had a significant difference(P＜0.05); According to clinical pathological stage,The positive rate of PCNAbetween IstageGroup,II stage Group,III stageGroupand N stage Group(42.86%,54.55%,80.00%,i00.00%.)Had a significant difference(P <0.05).The positive rates of PCNA increased with the increase in clinical stage,the increase in the degree of maignancy and the increase in infiltration(P＜0.05).In addition,PCNA protein expression of had no relation to the patients' age,sex,tumor size and location.(P＞0.05).3.The relationship between PTEN andPCNA protein expression in the gastric MALT lymphoma:The positive rate of PTEN and PCNA protein in gastric MALT lymphoma were 47.83%and 71.74%,and correlated with each other(P＜0.005),Pearson contingency of coefficient (p)=0.4879.Conclusion:The abnormal expression of PTEN and PCNA participate the malignant transformation process of gastricmucosa,The lower of PTEN expression and the higher of PCNA expression are closely Related to clinicalstage,the degree of maignancy and infiltration.The missing of PTEN expression in the promotion of the missing reduce the proliferation of cells,thereby increase PCNA expression.PTEN and PCNA joint detection in the gastric MALT lymphoma can be used as an objective indicators to determine the degree of malignancy,judgement the prognosis for clinical and guide Treatment.
