Objectives:To reseach the treatment effects and its mechanisms of puerarin on nonalcoholic fatty liver and atherosclerosis induced by high lipid diet in white rabbit.Materials and Methods:Thirty healthy male white rabbits were randomly divided into 5 groups:(1) Control group was fed with common forage;(2) Model group was fed with high lipid diet;(3) Treatment groups(PurⅠ,PurⅡand PurⅢ) of puerarin were constantly fed with high lipid forage and treated 2 weeks(once per day) at the end of the 5th week,which were intravenously administrated with 10,30 and 50mg/kg,respectively.At the end of the 0,5th and 7th weeks,the blood samples were collected by auricularis arterial puncture.Biochemical parameters of serum,such as total cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL-C), low density lipoprotein cholesterol(LDL-C),aspirate aminotransferase(AST),alanine aminotransferase(ALT),superoxide-dismutase(SOD) activity and malondialdehyde (MDA) were measured.The rabbit weight was weighed once per week to compare the average increased heavy.The rabbits were sacrificed at the end of the 7th week,the aorta and liver was removed en bloc.AS plaque areas of the aorta and liver index were calculated.The pathological changes of liver and aorta were observed with microscope.Results:The results were as follows.1.Biochemical parameter changes of serum.Before experiment,the contents of biochemical parameters in serum were physiological normal.After 5 weeks of high lipid forage administration,contents of TC,TG,LDL,ALT,AST,MDA activity were significantly increased compared with the control group(P<0.01),but the contents of SOD and HDL-C were significantly decreased compared with the control group (p<0.01 or p<0.05).After 7 weeks of high lipid forage administration,contents of TC,TG,LDL, HDL-C,ALT,AST,MDA and SOD changed as described below.(1) TC content in model and different Pur groups were more higher than control group(p<0.01 or p<0.05).PurⅠand PurⅢgroups were all lower than that in model group in the same period,there is no differences in PurⅡgroup compare with model group.(2) TG concentration in PurⅡand PurⅢgroups were significantly higher than control group(p<0.05),.there is no differences in PurⅠin the same experiment period(p>0.05),there is no differences in there Pur groups compare with model group.(3) HDL-C concentration in there Pur groups were significantly higher(P<0.01 or P<0.05) than that in control group and model group in the same experiment period. HDL-C concentration in PurⅢgroup is the most highest than other groups.(4) LDL-C concentration in the model group was significantly higher than control group.LDL-C concentration in three Pur groups were significantly lower (P<0.01) than that in control group and model group in the same experiment period.(5) AST concentration in the three Pur groups were significantly higher than that in control group.AST concentration in the three Pur groups were significantly lower (P<0.01) than that in model group in the same experiment period,there is no differences in PurⅠgroup compare with control group.(6) ALT concentration in the model group was significantly higher than that in control group.ALT concentration in the three Pur groups were significantly lower (P<0.01) than that in model group in the same experiment period,there is no differences in PurⅠand PurⅡgroups compare with control group,ALT concentration in PurⅢwas significantly higher than that in control group(P<0.01).(7) MDA concentration in the three Pur groups and in model group were significantly higher(P<0.01 or P<0.05) than that in control group in the same experiment period,MDA concentration in the three Pur groups were significantly lower than that in model group in the same experiment period.(8) SOD activity in three Pur groups were significantly higher(P<0.01) than that idn model group in the same experiment period,that there is no differences compare with control group.2.The degree of liver fatty degeneration,inflammatory activity and liver index. The degeneration degree of liver was the most serious in model group both macroscopy and microscopy.The degeneration degree of nonalcoholic fatty liver and liver inflammatory activity can be alleviated significantly by puerarin.There were no differences in different Pur group compare with model group about liver index.3.Pathological changes of aorta.AS plaque in the aorta was observed and protruded under macroscopy.Microscopic morphology showed that there was severe atherosclerotic lesion in the aortic arch in model group.The degree of atherosclerotic lesion in treatment groups was gradually alleviated as the Pur dosage increased.Conclusion:The results revealed that puerarin could resist formation of nonalcoholic fatty liver and antiatherosclerosis caused by experimental hyperlipidemia in rabbits.There were relationships between resistance effects of puerarin on nonalcoholic fatty liver and atherosclerosis formation and dosage administration.
