Object To study the effect of toremifene combined with Vinorelbine shore on the models of VX2 cancerous tissue transplantable subcutaneous tumor in rabbit and its mechanisms .Method 32 white rabbits were inoculated 2.0 ml suspension concentra(?)ing VX2 cancerous tissue on the right back leg of the animals respectively via subcutaneous injection to establish the models of VX2 cancerous tissue transplantable subcutaneous tumor. All the rabbits were randomly divided into four groups after the models were established. the control group, Toremifene group, Vinorelbine group and Combined-group (Toremifene the Vinorelbine );Toremifene group and Combined-group were admistrated Toremifene by oral (60 mg/kg) each day from the eighth day to fourteenth day respectively after inoculating VX2 cancerous tissue suspension ; Vinorelbine group and Combined-group were injected 2.5 mg vinorelbine on the twelfth day and 2.0 mg vinorelbine on the nineteenth day after inoculating VX2 cancerous tissue suspension respectively. All the VX2 cancerous tissue transplantable subcutaneous tumors were measured and compared the tumor growth rates once weekly .All animals were killed on the thirtieth day after inoculating VX2 cancerous tissue suspension. and observed the pathologic changes. The method of TUNEL dyeing were used to measure the apoptosis .Result Vinorelbine group and combined group showed evident effect of inhibiting the tumor growth , the inhibiting rate of the tumor growth were much higher that in control group((P<0.01) and the inhibiting rate of the tumor growth in the combined group was higer than that in Vinorelbine group(P<0.01) ; Toremifene group showed a certain inhibiting effects of tumor grow,but there was no evident difference compared with control group(P>0.05). lots of denaturization necrotic tumor cell can be seen in Vinorelbine group under HE dyeing, but less than that in the combined group. Vinorelbine group showed more apoptosis cell compared with Toremifene group under TUNEL dyeing, but showed less apoptosis cell compared with the combined group. In Toremifene group under HE dyeing tumor slightly necrocytosis could be seen under HE dyeing, tumor cell still were in row and connected closely and some apoptosis cell can be seen under TUNEL dyeing,ConclusionToremifene ( 60 mg/kg) combined with Vinorelbine could inhibit the growth of VX2 cancerous tissue transplantable subcutaneous tumor, show significant synergistic anti-tumor effect on rabbit-VX2 cancerous tissue transplantable subcutaneous tumor .Toremifene combined with Vinorelbine could induce VX2 cancerous tissue transplantable subcutaneous tumor cell capoptosis.
