Objectives:To investigate the distribution of human papillomavirus(HPV) genotypes and HPV L2 protein localization in uterine cervical lesions.Materials and Methods:72 cases of squamous cell carcinomas(SCC),28 cases of adenocarcinomas,and 196 cases of CIN-1 to CIN-3 were selected from Dept.of Pathology,Yanbian University Hospital,Yanbian Women's Hospital,and Yanbian Tumor Hospital in the period of 1996-2006,and HPV genotypes were detected by using oligonucleotide microarray.To study the significance of HPV L2 protein expression pattern,the monoclonal L2 antibody RG-1 was used to detect the L2 protein expression in uterine cervical lesions by immunohistochemistry.For the L2 protein localization study,HeLa cells were transfected by HPV L2 DNA plasmid,and then observed the L2 protein localization by immunofluorescence.Results:All the normal cervical epithelia are negative for HPV.The positive rate of high-risk HPV is 33.9%in CIN-1,51.6%in CIN-2,57.7%in CIN-3,91.7%in SCC, and 78.6%in adenocarcinoma.HPV 16 is the major type in all CIN and SCC cervical lesions.However,in cervical adenocarcinoma,HPV 18 is the most common type,and HPV 16 is the second type.Several cases of CIN-3,SCC and adenocarcinoma showed multi-types HPV,but none in CIN-1/2.Additionally,all the 20 cases of normal cervical epithelia are negative for RG-1 immunostaining.Likewise all the cases of CIN-3,sqaamous cell carcinomas,and adenocarcinomas are also negative for HPV L2 protein expression by RG-1 immunohistochemical staining,however,HPV16 positive CIN-1 and CIN-2 cases showed positive staining in the nucleus of the terminal differentiated epithelial cells.Also,L2 protein was co-localized with PML in the nucleus of HeLa cells which was transfected by HPV16 L2 DNA plasmid.Conclusions:HPV 16 is the type most frequently involved in the development of SCC of the cervix,and maybe helpful for the prediction of the development and progress of CIN-2/3,whereas both HPV 18 and 16 play a prominent role in the development of adenocarcinoma of the cervix.In addition,HPV L2 protein is strictly related to the terminal differentiated epithelium and co-localized with the PML in the nucleus of HeLa cervical cancer cells which transfected by L2 DNA plasmid,and the monoclonal HPV L2 antibody RG-1 is very helpful for the screening of HPV 16/18 infection status in CIN-1 and CIN-2.
