Objectives:To establish neoplasm transplantation models of melanoma in mice in order to study the therapy of melanoma.Methods:B16 melanoma cells in logarithmic growth phase were inoculated into BALB/c mice's back hypodermis and feet and C57BL/6 mice's back with different concentration to establish neoplasm transplantation of melanoma.The growth characteristics of transplanted melanoma in mice were observed and the tumor properties were identified by histopathology and immunohistochemistry.The relations between the number of B16 cells inoculated and tumorigenetic rate,and the influence of tumorigenesis for mice were observed.Results:1.Melanoma models in C57BL/6 mice.The tumorigenetic rate in inoculating with 2×10~5 B16 cells was near 100%.The time of melanoma emergence and mice's surviving time will be prolonged with the decrease of the density of inoculated cells, and differences exist among groups(P<0.05);however,no significant differences of tumor-bearing time exist among 3 groups(P>0.05).With B16 melanoma cells inoculated,emergence time of tumor appeared the earliest in high density group, during the first five days tumor grew the fastest,during the following eight days tumor grew smoothly,and then the growth slowed down,tumor-bearing mice's surviving time was the shortest.Emergence time of tumor appeared the latest in low density group,tumor grew smoothly at first,from the ninth to the fifteenth day tumor grew rapidly,and then at a low speed,tumor-beating mice's surviving time was the longest.The emergence time of tumor and mice's surviving time in the medium-sized group was between the two groups above,tumor grew slowly during the first seven days,in the following six days tumor grew rapidly,and then tumor grew slowly.No tumor disappearance emerged in all the three groups. 2.Melanoma models in BALB/c mice.Five days later,black spots and linear changes were gradually seen in BALB/c mice's back skin,feet or ankle,and then they developed to tubercles,ulceration,hemorrhage and death.The tumorigenetic rate in inoculating with 5×10~5 B16 cells was near 100%.Conclusions:1.The emergence of tumor and mice's surviving time are related to the number of inoculated cells.2.The models of melanoma in C57BL/6 mice have the advantages of a higher success rate.3.The models of melanoma in BALB/c mice compared with the other model indicated the former was easily to control and observe.
