Objective:To observe the changes of the activeness, shape, DNA and related functions of the vascular endothelial cells incubated by the serum of patients with blood stasis syndrome (BSS) associated with hypertension disease, and discuss the mechanism of the pathological changes and the role of these changes in the development of hypertension, BSS.Methods:The human umbilical vein endothelial cell (ECV-304) was intervened by the 15% serum of the patient of BSS associated with hypertension disease for 48h, the positive control group and negative control group was intervened by the 15% serum of the patient of non-blood stasis syndrome (NBSS) associated with hypertension disease, and healthy human, the control group was intervened by non-serum medium. Detected the following indicators:1. The activeness of the ECV-304 was determined by MTT assay.2. The damage of the DNA was determined by Single cell gel electro phoresis (SCGE).3. The expression of the mRNA of endothelin (ET), von Willebrand factor (vWF), thrombo-modulin (TM) was determined by the reverse transcriptase-Poly-merase chain reaction (RT-PCR).4. The content of malondialdehyde (MDA) in cell medium was measured by thlobarbiturie acid (TBA) reaction, and the activity of superoxide dismutase (SOD) measured by xanthosine Oxidase method.Results:1. Compared with the NBSS group, healthy group, the BSS group s activeness was decreased (P<0.05).2. The DNA of the BSS group and the NBSS group s cell showed the typical "comet" image . Selected some indicators which can reflect DNA damage we can see that: the DNA of BSS group, NBSS group s cell was significantly damaged. The damage of the BSS was more serious than the NBSS, but there was no difference between them.3. The expression of ET, vWF, TM mRNA of the BSS group, NBSS group s cells to the blank group was significantly increased (P<0.05), the expression of these three proteins mRNA of the BSS was more than the NBSS.4. Compared with the healthy group and the blank group, the BSS group and the NBSS group s cells released more MDA and SOD activeness decreased (P<0.05), but there was no difference between the BSS and the NBSS.Conclusion:The damage factors in the serum of patients with essential hypertension can cause endothelial dysfunction: the VEC s activeness was decreased, caused oxidative stress, cell s DNA was damaged, injury-related markers of endothelial expressed more. All of these play a very important role in the development of hypertension. The differences between the BSS group and the NBSS group are not very clear in the above-mentioned changes, but there is a trend that the BSS group changes more than the NBSS group, suggest that these pathological changes may related to the evolution from the NBSS to BSS.
