Effect Of Stichopus Japonicus Sekenka (SJAMP) On Caspase Expression Of Hela Cells

Objective To study the apoptosis of Hela cells induced by stichopus japonicus sekenka(SJAMP,)and its possible mechanism.Methods Hela Cells were treated by different concentrations of SJAMP in different time;The inhibitory effect on cell proliferation was assayed by MTT;the apoptosis was observed by electronic microscopy;The expression of caspase-3 and caspase-9 was assayed with reverse transcription-polymerase chain reaction(RT-PCR).Results SJAMP can suppress the growth of hela cells obviously in dose-dependent and time-dependent manners;morphology changes of cell apoptosis such as karyopyknosis and conglomeration were observed by electronic microscopy;RT-PCR assay showed the expression of Caspase-3 and caspase-9 increased after the exposure to different concentrations of SJAMP in different time.Conclision SJAMP could inhibit proliferation and induce apoptosis of Hela cells, Caspase-3 and caspase-9 may have an important rule in the regulation of cells apoptosis.
