Polygonatum sibiricum Red., which belongs to the Liliaceae family, is herbaceous perennial plant. P. sibiricum Red. has various pharmacological functions, for exemple, antiatherogenetic, anti-senility, anti-tumor, and so on. For further exploitation of P. sibiricum Red., the systematical study on the chemical components and pharmacology function of P. sibiricum Red.is necessary. Our study includes following parts:1. Weigh 25kg of P. sibiricum Red. grinded and passed the 20 mesh sieve. Ethanol extracts of P. sibiricum Red. was got by using alcohol reflux extraction. The extraction parameter was 70%(v/v), concentration of ethanol, 1:7(kg?L-1) ratio of solvent to raw material, 2 hours of reflux for 3 times. N.butanol extracts of P. sibiricum Red.was got after degreased by petroleum ether and extracted by n-butanol.N.butanol extracts of P. sibiricum Red. was coarsely separated by macroporous absorption resin(AB-8), the concentration of the eluent was water, 20%, 40%, 50%, 75% and 95% ethanol. The effluent was detect by TLC. According to color, Rf, and retention time, the effluent was divided into A, B, C, D, E, F, G, H and I. The steroidal sapogenin mainly distributed in G, H and I. These three parts were concentrated to remove fluorescent substance by using polyamide. Four compounds were finally isolated from these three parts by using ODS and MCI.2. Their chemical structure were elucidated by means of 1H-NMR,13C-NMR,135DEPT spectroscopic analyses. The four compounds are:3-O-β-D-glucopyranosyl(1→3)-β-D-glucopyranosyl(1→4)-[α-L-rhamnopyranosyl(1→2)] -β-D-glucopyranosyl-diosgenin3-O-α-L-rhamnopyranosyl(1→4)-[α-L-rhamnopyranosyl(1→2)]-β-D-glucopyranosyl-di-osgenin3-O-β-D-glucopyranosyl(1→4)-[α-L-rhamnopyranosyl(1→2)]-β-D-glucopyranosyl-dios-genin and diosgenin.CompoundsⅠand CompoundsⅢwere not reported from P. sibiricum Red..3. The antiatherogenetic effects of total saponins of P. sibiricum Red.(TSP) and compoundⅡon hypolipidemic mice were investigated. Compared with control, the value of triglycerides (TG) and total cholesterol (TC) of hypolipidemic mice were not decreased significantly by TSP, but decreased significantly by compoundⅡ(p<0.05).
