BackgroundsWound healing is a complex biological tissue repair process, which is entered from the wound fibroblast proliferation, differentiation and synthesis and secretion, extracellular matrix fibrosis, neovascularization and epidermal cell proliferation cover the wound such as re-epithelialization of the combined effects of the results. Wound healing can be roughly divided into three stages:①inflammatory response dissolved, removing necrotic tissue and exudation;②connective tissue cells and vascular endothelial cells swimming, multiplication, granulation tissue formation;③new connective tissue matrix deposition and alteration new organization, moderate inflammatory reaction is useful to the body in wound healing process and can generate protective immunity against infection, excessive inflammatory reaction can produce harmful pathological changes to the body. Fibroblast is a major component of the granulation tissue, wound partial undifferentiated mesenchymal cell different-tiation or static fibroblasts convert into fibroblast cells under stimulation of the injury, Fibroblast enter the wound and split under the growth factors and other chemokines. Fibroblast can synthesis and secretion collagen, extracellular metalloproteinases and their inhibitors. Fibroblast can lysis extracellular matrix, and participate in the reconstruction.abundant blood capillary can enrich granulation tissue and improve microcirculation for the organization to provide rich oxygen and nutrients, thus helping wound healing. These three stages are complementary and mutually linked, each phase are adjusted delicately by the body. Wound healing researching has been a clinical focus, senior citizens, poor nutrition, infection and poor local blood circulation, and so on, caused delay healing, disunion, delayed hospitalized time increase the economic, physical and psychological burden to patients. In recent years, many cell growth factors can be used to accelerate organizations as drug rehabilitation and repair bad state of downturn with the clinical application of molecular biology, but still be greatly restricted beacause of growth factor leading to the application of hypertrophic scar, cholesteatoma and malignant cells. In tissue engineering, the better is composite skin tissue engineering, but there will be different function in the tissue-engineered skin and normal skin. Stem cell technology obtain a number of accomplishments, but is still at the preliminary stage, a large number of issues must be resolved before clinically utilized. At present, many herbs have anti-infective, wound exudate reduction, accelerated epithelial, vascular growth function, and reduce scar tissue proliferation, and other role. However, there will be more need in-depth study and practice of development to promoting more efficient drugs in line with international standards and requirements. How to accelerate the wound healing clinical studies are now one of the hot spots.Ecdysterone (ecdysterone, EDS) is known asβ-molting hormone and insect metamorphosis hormone in animals, which stimulates insect leather cell division to creat new skin, result insect metamorphosis, and regulate insect metabolism. Ecdysterone can affect glucose metabolism, and promote lipid metabolism, immune regulation, affect the central nervous system. In recent years, many reporte in the literature discoverd that it can stimulate secretion of vascular endothelial in acute myocardial infarction, and promote angiogenesis the establishment of collateral circulation, capillary density in the ischemic region. It also can promote wound healing. Plant chemical studies showed that the ecdysterone is very wide distribution in the Flora.The studying of ecdysterone made to ointment preparation and the infuence of wound healing on rabbit provide a basis for the clinical application of trauma wounds and promoting more developed new types of agents drugs. ObjectiveTo observe the wound healing and capillary formation effect in the raw surface in rabbits' back of ecdysterone ointment.Mothods1. Monostearin 35g, Octadecanoic 140g, White Vaseline 70g, Azone 20ml, and Hydroxy ethyl benzene 1g is heated to 90 degrees by water-bath until they were melting at oil phase; Purified Water 580ml, Borax 4g, Sodium lauryl sulfate 4g, Riethanolamine 15ml, Glycerol 160ml and Allantoicase 3g is heated to 90 degrees by water-bath until they were melting at aqueous phase; ointment matrix was maded while the oil phase mixture was joined in the water phase mixture slowly. Ecdysterone ointment is maded in 25g·kg-1 concentration by taking the ecdysterone dissolved in ethanol solution adding in a certain amount of matrix-stirring when it was to be agitated.2. There were 3 raw surfaces trauma model everyone in the 24 rabbits, while the 3 raw surface was treated by 2.5% ecdysterone paste, Yunnan white powder, paste substrate respectively, the curative effect was to be evaluated by the rate of restitution in the 12 rabbits on 4, 8, 12 days after operation, derive granulation tissue from the other 12 rabbits on 4, 8, 12 days after operation, the curative effect was to be evaluated through capillary density in the raw surface by the immunohistochemical staining of vascular endothelial cell marker observation making use of the neogenesis granulation tissue of the raw surface.Result1. Basic observe the wounds of 3 group are swelling and exudate, but no obvious infection on 4d after operation, the swelling and exudate are lighter in the wounds deal by ecdysterone intment than the wounds deal by thers. The granulation tissue hyperplasia is not obvious in 3groups. After 8d, the woundsare moist and no obvious infection in ecdysterone ointment group and Yunnan Baiyao group wound, the contraction of wound surface and subcutaneous tissue are obvious, and the proliferation of granulation tissue is obvious, the wounds of matrix control group are still swelled and exuded, the contraction of wound surface and subcutaneous tissue are bad than that of ecdysterone group and Yunnan Baiyao group. After 12d, the wounds of ecdysterone ointment group and Yunnan Baiyao group are dry, surface scared, nearly complete coalesced, but part of woungs of the control group Matrix are still exuded, the wound healing is bad than that of ecdysterone ointment group and Yunnan Baiyao group.2. Comparison of healing rate There isn't significant group differences in the healing rates among the 3 groups on 4 days after operation (P>0.05), while the healing rate of the ecdysterone ointment group and the Yunnan Baiyao group is better than that of the control group on 8 and 12 days after operation (P<0.05), and there isn't signify-cant group difference between the ecdysterone ointment group and the Yunnan Baiyao group (P>0.05).3. HE staining observe The three groups are wound exudate and congestive, the granulation tissue hyperplasia is not evident 4 days after operation, 8 days after operation, the capillary and epithelial hyperplasia is not obvious in the control group. There are a large number of capillaries, fibroblast proliferation, some infiltration of inflammatory cells, collagen and some epithelial cell proliferation in Ecdsterone ointment group. There are also a large number capillaries proliferation of fibroblasts, a little of the infiltration of inflammatory cells in Yunnan Baiyao group. 12 days after operation, there are still some defects, a large number of inflammatory cells exudative, and only a little granulation tissue, a small amount of epithelial cell in control group, three are much granulation tissue, and some inflammatory cell infiltration between the dermis and epidermis connect in the ecdysterone ointment and Yunnan Baiyao groupe; the collagen formation,epithelial cells and fibroblasts cell proliferation is obvious.4. Comparison of blood capillary density There isn't significant group differences in the blood capillary density among the 3 groups on 4 days after operation (P>0.05), while the blood capillary density of the ecdysterone ointment group and the Yunnan Baiyao group is better than that of the control group on 8 and 12 days after operation (P<0.05), and there isn't significant group difference between the ecdysterone ointment group and the Yunnan Baiyao group (P<0.05). ConclusionThere is prominent effect of promoting the wound healing and capillary formation in raw surfaces by ecdysterone ointment.
