Objectve Hepatocellular carcinoma is a kind of malignant tumor which has high morbility in worldwide. early detection is of great importance for diagnosis and prognosis. Now, none of tumor marker could detect the all kinds of hepatoma. We hope to study the expression of Receptor binding cancer antigen expressed on SiSo cells (RCAS1) in human hepatocellular carcinoma (HCC) and its relationship between the expressions of RCAS1 and the clinic pathological characterizations of patients with HCC.Methods The expressions of RACS1mRNA in samples of hepatocellular carcinomas and Corresponding adjacent noncancerous tissues were detected by real-time fluorescence quantitative PCR(RFQ-PCR). Samples from 37 surgically resected and microscopically diagnosed HCCs were collected in the Department of surgery, Union Hospital from March,2005 to December,2006, Total RNA was extracted and The concentration of RNA was tested by ultraviolet spectrophotometer, then conserved at -80℃.In iced bath. AMV reverse transcriptase was added in RNA, cDNA was synthsised in certain experiment condition, Real-time fluorescent polymerase chain reaction was used to monitor the cycle threshold value and analyze the expressive production of target mRNA. The clinicopathological data of these 37 patients were analyzed.Results 1. RCAS1mRNA expression in hepatocellular carcinoma tissues was much higher than that in the adjacent noncancerous tissues(P<0.05).2. The expression of RACS1mRNA in HCC was not correlated with HBsAg, tumor size and serum AFP, but significantly correlated to histological differentiation and TNM stage(P<0.05).Conclusions 1. The expression of RCAS1 in hepatocellular carcinoma tissues was much higher than that in the adjacent noncancerous tissues. 2. RCAS1 may be a new predictive marker for HCC and is of potential value in judging invasion and progression of HCC.3. The real-time PCR assay with SYBR Green I via Lightcycler instrument is specific and sensitive than before in studying the expression of mRNA.
