Objective To discuss the mechanisms of the radiosensitization of paclitaxel to the non-small cell lung carcinoma cell line SPC-A1. Methods Examining the cell toxicity of paclitaxel to the SPC-A1 cell with MTT; the clone forming test is the used to examine the radiosensitization of paclitaxel to SPC-A1 and the enfluence of paclitaxel to sublethal damage repair; the single cell gel electrophoresis is used to examine the enfluence to the DNA repair. Results The IC50 of paclitaxel to SPC-A1 is 50 nmol. The SER of paclitaxel to SPC-A1 is 1.42. In the test of fractionated irradiation, the ratio of the survival rate of the interval of 5hours to that of 0 hour is 1.96 in the single radiation group, wheras the ratio is 1.53 in the group of paclitaxel. There isn't significance difference in tail moment (TM) between the two groups at 0 min after radiation(P>0.05), there are significance differences in TM between the two groups at 15,30,60min after radiation(P<0.05). Conclusion paclitaxel can inhibit the repair of the non-small cell lung carcinoma cell line SPC-A1 damage resulted from the in vitro radiation.
