Glucosinolats are a kind of indican, which widely exist in the roots, caudexes and fruits of Cruciferae. Especially, the plant family of Brassica, including cabbage, broccoli and Brussels sprouts, has a high glucosinolate content. The hydrolytic products of glucosinolates have been shown to partly prevent carcinogenesis in several experiments. The anticarcinogenic effects of hydrolytic products were proved in vivo and in vitro, and the mechanism was also primarily studied in this thesis. Then researches on the distribution of glucosinolates in coles and process influence were presented. At last, total glucosinolats were extracted from coles. The purpose of these studies was to provide theoretical support for the anticarcinogecity of cruciferous and screen medicine materials from coles.(1) Systematically pharmic researches on three hydrolytic products, which wereβ-phenylethyl isothiocyanate (PEITC), Benzyl isothiocyanate (BITC) and Indole-3-carbinol (I3C), had been done by setting up the mice model of transplantable S180 tumour in vivo. The inhibition effect was evaluated by the tumor weight of the mice bearing tumor, the tumor inhibition rate and the spleen and thymus index. The results showed the high dosage had the significant anticancer activity to S180 cells, and the declining of spleen and thymus index indicated that PEITC, BITC and I3C had the effect of immunosuppressant in vivo.The anticancer effect of in vitro was studied by MTT assay about cultured HeLa cells. It was obtained that the cell proliferation was obviously inhibited by them in vitro. And the inhibitive effect was better than Cyclophosvnamide.(2) Observed the influence of PEITC, BITC and I3C on MCF-7 cells cycle and apoptosis. The results showed they could regulate cell cycle progression and significantly induce apoptotic cell death of MCF-7 cells, primarily in S phase. The modality of MCF-7 cells were also observed by AO/EB fluorescent coloration.(3) Determined the glucosinolats content in rapeseeds, laminaes and caudexes from nine breeds by palladium chloride spectrophotometry analysis. Screened the breed which had the highest content in every part especially in the rapeseeds. Then treated the samples in different temperatures and found that myrosinase was inactive at the temperature above eighty centigrade.(4) The total glucosinolats were extracted from rapeseeds by the methods of acid alumina chromatogram and DEAE-Sephadex A-25 resin chromatogram. Then the extractions were analysised by thin-layer chromatogram. And the results showed that three hydrolyzable products were contained in the extractions. Their contents were determined by high-performance liquid chromatography, and results showed their contents were 3.33μg, 1.38μg and 1.85μg/g rapeseeds. It was indicated that coles had the potential pharmic value of development. These researches found a new way for advancing the useful value of cole resources.
