Object to investigate the feasibility of tissue engineering anterior cruciate ligament constructed by silk scaffold which use the method and principle of tissue engineering.Method 1. preparation of allogeneic tendon and silk scaffold then measurement of material properties; 2. culture and identification of mice marrow mesenchymal stem cells; 3. the MSCs adhered on the silk scaffold were observed by the scanning electron micrograph(SEM).Result 1. The main parameters (Max-load, Max-strok,elastic) were not significantly different between sample A group and sample B group (P>0.05), but the Max-load and elastic of silk scaffold were prefer to allogeneic tendon(P<0.01). 2. The cultured cells were characterized by spindle shape appearance and positive expression of CD44, but CD34 was negative. 3. The SEM showed the silk scaffold had smooth surface and the MSCs had adhered the matrix fastly.Conclusion A three dimensional silk scaffold has good tissue and mechanical compatibility, It can be used to support MSCs in vitro, and may become the ideal biomaterial for constructing a tissue engineering ACL if the mechanical properties can be improved.
