Objective:To evaluate the clinical significance of serum IL-17 and IL-6 in patients with kidney transplantation and the significance of MIF expression in allotransplantation model mice.Methods:Serum IL-17 and IL-6 were detected in 30 patients with renal transplantation before and post transplantation on day 7,14,21 and 28 with ELISA and radioimmunoassay MIF expression was detected by RT-PCR in model mice of allotransplantation.Recombinant MIF was radio-iodinated with Na125I,then 125I-rMIF was isolated by Sephadex G25.The stability,immune specificity, and its biodistribution in the mice were studied.Results:There were no significant difference existed in the levels of serum IL-17 and IL-6 in pre-transplantation patients compared to those of control group respectively(p>0.05).After operation,the levels of serum IL-17 on day 7 and day 14 in patients were significantly higher than those of pre-operatively and were the highest on day 21(p<0.05),then dropped to pre-operative values on day 28.The serum IL-6 levels increased significantly than those of pre-operatively from day 7 until day 28(p<0.05),and decreased slower than the serum IL-17 levels.MIF expression was higher on the day 14 which is the top point of rejection.The labeling rate of 125I-rMIF was 96.5%.It was stable within 48 h at room temperature.The biodistribution results showed that the 125I-rMIF was metabolized by the liver,the radioactivity clearance mainly happened in the kidney and the speed of the blood clearance is rapid.Conclusion:1 The levels of serum IL-17 and IL-6 may reflect status of immune response in kidney allograft recipient,which are useful markers for identifying acute rejection after kidney transplantation,2 MIF expressions were increased during allograft rejection,so 125I-MIF may be used to detect the acute allorejection.
