Objective: The expression of integrinαvβ3 receptor in turner cells and vascular endothelial cell with 99mTc labelled RGD peptides on animal model was studied in the project.Method: The expression of integrinαvβ3 receptor in normal cell line and four tumor cell lines were detected with the cell fluorescence labelling techenique. ?(99m)Tc was labelled on RGD peptides with HYNIC. A model of mouse of H22 hepatoma cell was established. The mice were injected tracer via caudal vein and were scanned with SPECT Uptake rate of tumor cells for 99mTc-RGD wascalculated by a semiquantitative method of ROI.Result: Integrinαvβ3 receptor expressed rarely on normal cells but strongly on turner cells. To compare with 99mTc-HYNIC-RGD (non-NA),99mTc-HYNIC-RGD (EDDA) and 99mTc-HYNIC-RGD (NA) ware less accumulated and faster cleared in liver, was higher radiation and performed better image on tumor detecting, and got higher ratio of target to background on SPECT imaging. Time of the maximum count ratio of tumor to background was 3 hours after injection.Conclusion: Mouse with H22 hepatoma cells took 99mTc-HYNIC-RGD evidently. The best time of SPECT imaging was 3 hours after injection. 99mTc-HYNIC-RGD is one kind of potential tracer of diagnosis and targeting therapy of tumor.
