Effects Of Soothing Liver, Invigorating Spleen Recipes On Expression Of PI3K P85a Protein In Rats With NAFLD

Objective:To investigate the effect of soothing liver,invigorating spleen recipes on insulin resistant,serum lipid,liver function,liver histopathology and expression of PI3K p85a of liver cell in rats with non-alcoholic fatty liver,and to approach its possible mechanism on treating NAFLD.Methods:Seventy-six male SD rats were randomly assigned to normal group,model group, soothing liver group,invigorating spleen group,complex recipes group(soothing liver recipes and invigorating spleen recipes),sanqi group,high-fat diet control group,10 rats in each group.The normal group was fed with normal diet,the other groups was fed with high-fat diet(83%standard diet,10%pig fats,5%suger,1.5%cholesterol and 0.5%bile salt were added).By the end of 12^th weeks,soothing liver group received gastric perfusion of chai hu shu gan san,invigorating spleen group received gastric perfusion of shen ling bai shu san,complex recipes group received gastric perfusion of chai hu shu gan san and shen ling bai shu san,sanqi group received gastric perfusion of san qi zhi gan wan,while rats in other three groups received gastric perfusion of distilled water at the same volume.The model group was still fed with high fat diet while the other groups with normal diet.All rats were executed at the 20^th week of feeding.The degree of insulin-resistant was measured with homeostasis model assessment.The ALT,AST,TG,TC,FFA, LDL-C,HDL-C,GLU,ApoB100 were measured by biochemical methods,and insulin was measured by radioimmunoassay,The assessment of hepatic steatosis,inflammatory activity in all rats were scored according to criteria under light microscope,protein expression of PI3K p85a were measured by immuhistochemisty.Results:1.Compared with normal group the contents of serum TC,TG,LDL-C,ALT,AST,FFA, GLUand ApoB100 in model group increased(P＜0.01 or P＜0.05),serum HDL-C decreased(P＜0.01).Histological changes demonstrated that rats fed the high-fat diet developed diffuse macrovesicular steatosis,lobular inflammatory cell infiltration.2.The expression of p85a in hepatic cell were seldom in the normal control,its expression in model group were significantly increased(P＜0.01).3.Compared with the model group,serum lipid,blood glucose,liver function and insulin resistance were inproved in soothing liver group,invigorating spleen group,complex recipes group,sanqi group,high-fat diet control group(P＜0.05),the expression of PI3K p85a in liver cell were decreased(P＜0.05).Compared with the high-fat diet control group,the difference of serum lipid,blood glucose and liver function in rats treated with soothing liver and invigorating spleen recipes associated diet treatment were of no statistical significance(P＞0.05),but the difference of insulin resistance,hepatic steatosis and inflammation activity and the expression of PI3K p85a in liver cell were of statistical significance(P＜0.05).Conclusions:1.NAFLD insulin resistance model was successfully developed in rats fed a high-fat diet,which had a mimic pathological and metabolic changes of fatty liver in patients.2.High-fat diet resulted in a significant increase in protein expression of PI3K p85a of hepatic insulin signaling transduction passway in rats liver cell,and the distribution of protein expression was in concordance with hepatic cell steatosis,which might be one of the molecular mechanisms of insulin resistance induced by high-fat diet,to cause lipid metabolic disturbance all over the body,thus fatty liver was formation.3.Stagnation of liver-QI with deficiency of the spleen and spit and blood stasis in the base of it were main pathogenesis of NAFLD,the production of insulin resistance was in correlation with closely stagnation of liver-QI,deficiency of the spleen and spit.Thus,improving insulin resistance by using soothing liver and invigorating spleen recipes could treat fatty liver.4.In earlier period of NAFLD,dietary control method can improve serum lipid,blood glucose and liver function.but soothing liver and invigorating spleen recipes associated with diet treatment can obviously improve insulin resistance and hepatic tissue pathology.By reducing maybe expression of PI3K p85a of liver cell,the method of soothing liver and invigorating spleen can ameliorate insulin sensitivity,reduce blood triglycerides,improved and ameliorate the hepatocytes fatty degeneration to treating effects on NAFLD.
