ObjectiveTo observe the effects of tanshinoneⅡA(TanⅡA),astragalus polysaccharide(APS) and their combination on the activeness,shape,internal secretion and cells'intracellular free calcium of human umbilical vein endothelial cell lines(ECV-304) injuryed by serum of patients with hypertension disease.To investigate Traditional Chinese Medicine of Yiqihuoxue monomer and their combination on the protection of endothelial cells,to supply the test foundation to Traditional Chinese Medicine of Yiqihuoxue on preventing and treating hypertension,protecting endothelial cells.MethodsExperiments were divided into five groups:the control group,the model group,TanⅡA groups,APS groups,the mixed groups.Cells'activenesss was assayed by MTT,cells'shape was observed by the inverse phase contrast microscope and HE dyeing,the nitric oxide(NO) excreted by cells was assayed by nitric acid deoxidizing enzyme method,and the endothelin(ET) was assayed by non-balance method,cells'intracellular free calcium([Ca2+]i) as assayed by laser scanning confocal microscope(LSCM).Results1.After the serum acted on cells for 24h,the cells'activeness was lower in the model group than the control group(P<0.05);when TanⅡA(40,20,10μg/ml),APS(640,320,160μg/ml) groups acted on cells,the cells' activeness was higher than the model group(P<0.05or P<0.01); when the mixed groups of TanⅡA,APS groups acted on cells,the cells' activeness was higher than the model group(P<0.01),than the single using TanⅡA or APS(P<0.05);2.The difference of shape was distinct between the modle group and the control group,the shape of the medicine groups close to normal cells'shape with the increase in medicine concentrations;3.NO secreted was lower in the model group than in the control group(P<0.01);ET secreted was higher in the model group than in the control group(P<0.01);NO secreted was higher in the TanⅡA,APS groups and the mixed groups than the model group(P<0.05orP<0.01),ET secreted was lower in the TanⅡA,APS groups and the mixed groups than the model group(P<0.05orP<0.01);and NO,ET secreted was distinct difference between the groups of TanⅡA,APS and corresponding the mixed groups(P<0.05);4.Fluorescence intensity(FI) was higher in the model group than the control group(P<0.01);FI was lower in TanⅡA,APS groups and the mixed groups than the model group(P<0.05),and FI was lower in the mixed groups than the single using TanⅡA or APS(P<0.05).Conclusions1.Model cells have the decrease in activeness,the change of shape and the injury of function;2.TanⅡA,APS and their combination can inhibit the decrease in cells' activeness,repair the changed shape and protect the function of damaged cells;there is a much better effect in the combination of TanⅡA,APS than the single using TanⅡA or APS.
